Melatonin is the principal neuroendocrinologic hormone secreted by the pineal gland, and is of the physiologic modulation in mood, sleep, sexual behavior, reproductive alterations, immunologic function, and circadian rhythms. As a free radical scavenger, melatonin can take protective effective on the central nerve system and may be important in slowing the processes of ageing. The melatonin coupled to G protein and mediated by the specific high affinity receptor localized on membrane. Two different 0 proteins coupled to the melatonin receptors have been described as Me11A and MellB.One is sensitive to pertussis toxin and the other sensitive to cholera toxin. Melatonin receptor regulates several second messengers:cAMP, cGMP, diacylglycerol, inositol trisphosphate, arachidonic acid In this study, using the method of JHC and ISH we manage to resolve some questions as this: 1 .Are there melatonin receptors in the embryo brain? 2. Which position do the melatonin receptors exist in the embryo brain? The final aim is to supply some laboratory data for the study of the mechanism that how the melatonin regulate the brain function. Main methods and results: 1 .The brain tissues were sliced and melatonin receptor were detected using immunohistochemistry method. The results show that brown granules deposited in cerebellum or hypothalamus cytoplasma and membrane treated by the antibody of MT1 receptor.When treated by the antibody of MT2 receptor, brown granules deposited in the cytoplasma and membrane of hypothalamus, occipital and medulla cells.2. Hybridization and dyeing were taken using MT1 and MT2 probe respectively. Theth--:4rs k$tCtmrvra}*t+N4F EjJ:tetresults show that, using the MTl probe, the plasma of the cell ofhypothalamus cell are positively dyed, using the MT2 probe,theplasma of the cell of hypothalamus, occipital, medulla cells arepositively dyed. It indicate that MTl and MT2 exist in the embryob r a i n.Conclusion: Two kinds of melatonin receptors, MT l and MT2exist on the membrane and cytoplasma of brain cells. These indicatethat the human embryo brain is a target organ of melatonin. It can acton the brain directly and regulate the brain function.
