Preliminary study on autoantibodies in sera from multiple myeloma patients and a novel autoantigen with MW 42.7KDMaster candidate Li HongmeiSupervisorsZhong Renqian Kong XiantaoChangzheng Hospital, Second Military Medical UniversityAbstractThis study is to explore autoantibodies and their target antigens in patients with multiple myeloma(MM). The positive rate was 41.04 percent in 453 MM cases by the use of indirect immunofluorescence and Western blotting. The dynamic changes of autoantibodies and their fluorescent paterns could be found in the course of MM development. With Western blotting, cellular nuclear extract from Hep-2 cells was reacted with 40 sera with strong ANA positive. The results showed that all sera react with a novel band with 42.7 KD which was different from all known autoantigens. This novel protein was isolated and purified with SDS-PAGE and two dimentional electrophoresis and sequenced with automated fluid phase sequencer. The N-terminal amino acid sequence was as following: His-Leu-Ser-His-Ltu-Val-Glu-Ala-Tya-Gln-. There is no same amino acid sequence in the data from GenBank, but 50% and 70% homologies could be respectively found in nuclear cap binding protein (human) and human cathepsin G precusor.
