Objective: In order to illustrate the clinic significance of megakaryocytic progenitors in some blood diseases. Methods: <1>By using the collagen semisolid culture method, we observed the traits of colony forming unit-megakaryocyte(CFU—MK) and burst forming unit-megakaryocyte(BFU—MK) in 18 patients with idiopathic thrombocytopenic purpura(ITP), 10 with aplastic anemia(AA), 10 with myelodysplastic syndrome(MDS) , 7 with acute leukemia(Al) and 12 normal controlls. <2> By using the APAAP method, we still observed the expression of T-cell subgroup in the peripheral blood cells in ITP and AA patients. Result: In AA group ,the colonies(10.60±14.51 ) of CFU-MK,BFU-MK were less than that of the normal controlls(49.42±15.84 )(p<0.001),8 patients(80%) were strikingly decreased,but 2 patients(20%) were normal. In ITP group, the colonies(46.67±19.43)of CFU-MK,BFU-MK were not significantly different from the normal controlls(P>0.05), 4 patients(22%) were decreased, and 14 patients(78%) were normal. In MDS group, the colonies(12.40±15.36) of CFU-MK,BFU-MK were less than that of the normal controlls(p<0.001), 8 patients(80%) were strikingly decreased,but 2 patients(20%) were normal. In AL group, the colonies(19.29±28.23) of CFU-MK,BFU-MK were less than that of the normal controlls(p<0.001), 5 patients(5/7) were strikingly decreased,but 2 patients(2/7) were normal. Conclusions: The colonies of CFU-MK,BFU-MK were decreased in many of AA,MDS,AL patients. The colonies of CFU-MK,BFU-MK in ITP patients were normal or decreased, maybe be related to immune disorders. In vitro culture of megakaryocytic progenitors can help us study the pathogenesis,therapies and prognoses in ITP, AA,MDS and AL patients.
