Study of the Gas-R and Signal Conduction in SpleenDeficiency Rats and the Effect of Chinese HerbZHANG GengshuiSupervisor: Prof WANG Rujun, Prof ZHOU FushenGuangzhou University of Traditional Chinese Medicine (Guangzhou, P.R.C. 510405)Abstract:BACKGROUND AND AIM Gastrin receptor(Gas-R) and the signal conduction are hot themes in gastroenterology overseas. Since 1980's, the relation of gatrin and spleen deficiency in TCM was studied by domestic experts. Untilled 1990's the studies was not progressed to the level of GasR and its signal conduction. The studies of Nie and Zhao foreshow that the Gas-R and its signal conduction were in a low metabolic condition in spleen deficiency syndrome. In the following study developed from these ones, the gastrin receptor binding sites, the level of calmodulin (CaM) .and the activity of calcium!calmodulin dependent protein kinase (Ca2~/CaM-PK) were observed. The effects of Huangqi on which were investigated at same time. The aim was to discover the relationship between spleen deficiency and gastrin receptor and its signal conduction.METHODS ~The spleen deficiency models were made by administrating Dahuang and reserpine. The rats were treated by Huangqi in control groups. ~Radio1igand binding assay wasestablished in screening gastrin receptor of parietal cells. ~~PDE method was used to observe calmoculin (CaM). @ Calciumlcalmodulin dependent protein kinase (Ca2~/CaM-PK) activity is in proportion to the substrate phosphorylation, so the activity was evaluated through measured the phosphorylation of substrate which was designed to bind to phosphocellulose.RESULTS tD The symptoms of the rats in spleen deficiency model groups were similar to which of the patients in clinic. These symptoms were markedly ameliorated after treated by Huangqi. e~Z~I In spleen deficiency model rats caused by reserpine, the gastrin receptor binding sites of parietal cells decreased markedly (271.2?1.8). Which were lower than that in normal group (619.1+304.5 P
