Purposes:To observe whether ionizing radiation could induce up-regulation of Fas receptor expression and apoptosis in diffuse type stomach carcinoma. To investigate the relationship among ionizing radiation, apoptosis and the expression of Fas in stomach carcinoma.Methods:Firstly, establish the experimental model of SGC?90l cell lines, diffuse type stomach carcinoma, orthotopically implanted in nude mice. Then 21 model mice were randomized into three groups averagely, control group (A group) and two irradiation groups(B group and C group, executed at 24 hours and 48 hours respectively after irradiation). The mice of B group and C group were irradiated with 6MV X-rays at 2OGy. By using the methods of TUNEL and immunohistochemieal staining, the changes of apoptosis index and Fas expression in tumor tissues were examined.Results:(1)The spontaneous apoptosis index (Al) of tumor tissues was significantly lower than that of mucosa tissues(P0.05).(3) It indicates that irradiation can up-regulating apoptosis in diffuse type stomach carcinoma cells, which have a time-dependent effect. The Fas LI of tumor tissues increased after irradiation compared with control group(P<0.05). It indicates irradiation can induce the up-regulating of Fas expression. (4)The changes of AT and Fas LI in all groups having similar tendency shown aIIIpositive correlation(PKO. 01).Conclusions:(1)The apoptosis of diffuse type stomach carcinoma is seriously restrained. (2)Ionizing radiation can induce apoptosis in diffuse type stomach carcinoma. (3)Ionizing radiation can induce up-regulating of Fas expression in diffuse type stomach carcinoma. (4) The apoptosis induced by irradiation maybe depend on the up?regulating of Fas after irradiation.
