ABSTRACT Objective: 1. To establish an allogenic limbal transplantation model with 3H-TDR stably labeled in New Zealand rabbits. Grafts were labeled with 3H-TDR and their 13 counts were determined after 3 month to track grafts survival. 2. Limbal transplantation in earlier period of corneal alkali burns was compared with limbal transplantation in later period to evaluate its efficacy in case of severe corneal alkali burns and provide evidences on theory and practice for the treatment of corneal alkali burns in acute stage of corneal alkali burns. Methods: 30 New Zealand white rabbits were divided into three groups: contrast group(CONs), limbal transplantation in earlier period group(ETG) and limbal transplantation in later period grotip(LTG). Limbal epidermis in donor rabbits?left eyes were labeled with 3H-TDR. Then receptor rabbits? right eyes were burned with iN NaQH. After burns, limbal transplantation was performed immediately in ETG and limbal transplantation was performed after 1 month in LTG. After surgery, the information of bilateral allografts on color, clarity, dissolution, exclusion, suture, hyperemia, scar was recorded at every times; PAS and HE staining were performed on both eyes and 31-I-TDR B radiation counts were determined. All gained data were analyzed with SPSS 10.0. Results: 1 .The allogenic limbal transplantatin model with stable label was established successfully. After 3 months, 3H-TDR B radiation counts in receptor rabbits of ETG were 702.3 ?40.O2CPM and that of LTG were 690.4 ? 47.69CPM. They were all higher than negative compared groups (P<0.05) . 2. After 3 months, the movment of new capillary invading limbal and centrality were 8.9?.37 days and 12.4?.41 days in CONs; 17.7?.21 days and 22.4?.02 days in ETG; 15.4 ?.36 days and 19.3 ?.26 days in LTG. Both ETG and LTG were lower than CONs (P<0.05) . The corneal epithelium heal uptime was 21?.5 days in CONs; 13.9?.37 days in ETG and 14.7?1.33 -4- days in LTG. Exps were lower than CONs (P
