Isolation and Molecular Characteristics Of Borreliaburgdorferi senso lato Of GuangxiM.Sc Candidature :Min LiuSupervisor:Prof. Shusheng Wang ABSREACTObjective To isolate lyme disease spirochetes from their host梤at in Yachang forest of Guangxi Province; and describe their molecular characteristics ; identi1~?their genospecies. Methods The rat's urinary bladder was inoculated to the BSK- II. After incubating for 7-14 days, the Borrelia burgdorferi can be observed in dark -field microscope. The antigen slides which were made of strains have reaction with monoclonal antibodies H5332,H683 1 ,H9724,respectively. Using rrf-rrl intergenic region primers of B.b.s.s the GXLD-4,9, 18 have intergenic spacer amplified. The rrf-rrl spacer amplication was cleavaged by DraI and MseI restriction enzymes .The restriction enzyme patterns could be divided into at least 10 different species or genomic groups. Results From these rat, six isolates were obtained which were named GXLD-4,8,9,16, 18,20, respectively. The positive ratio of isolation is 40%(6/1 5). The monoclonal antibodies reaction of GXLD-4 present H5332(+), H683 1(+), H9724(-); and GXLD- 18 H5332(-),H683 1 (-),H9724(+).The rrf-rrl space amplication restriction patterns of these isolates were non-typical and different. Conclusion It is the first report that Borrelia burgdorferi is isolated from Guangxi Province. The strain restriction fragement length phlymorbism of rrf(5s)-rrl(23s) shows difference and non-typical.
