Natural autoantibodies (NAA) refer to antibodies that are present in the serum of healthy individuals and can bind to one or more self-antigens in the absence of deliberate immunization. NAA are encoded by germline genes and belong to IgM~ IgG and IgA isotypes. NAA are polyreactive, and affinity of NAA to self-antigen varies in an abroad range. As V region-mediated networks are established by NAA, it is of great importance for NAA to act in immune homeostasis. NAA also play important roles in first line defense against infection anti-inflammatory activity~ anti-tumor surveillance-, antigen presentation to T cells and so on. Existing of NAA contradicts the classical theory of 揷lone selection? NAA not only have theoretical importance, but also are important clinically. Changes of NAA in quality and/or quantity are always involved in many diseases. As a member of the family of NAA, anti-keratin autoantibodies (AK auto Abs) are present in the serum of human and most of animals, playing an important role in many physiological and pathological -7- conditions of skin. After many years of research, we have proved the ubiquitous existence of AK auto Abs, and purified AK auto Abs from sera of healthy individuals by means of affinity chromatography. Basic characters of AK auto Abs have been learned. And studies also indicate that AK auto Abs are helpful to the recovery of animal model of psoriasis and contact dermatitis. Hybridoma secreting monoclonal AK auto Abs are made. And anti-keratin antibody is also cloned from phage display antibody library. But the research on the reactive pattern of AK auto Abs to keratin in different individuals and their development, changing with disease have not been conducted. The results of research on these subjects not only can give people complete understand of the physiological and pathological role of AK auto Abs, but also can help us to analyze many phenomenon, such as the occurrence and adjustment of AK auto Abs and their mechanism. This study tried to explain the above questions. Firstly, keratin purified from callus of healthy human hands and foot being used as antigen, ELISA and Westernblot were applied to analyze the immunoreactivity of AK auto Abs of IgG and 1gM type in the sera of health individuals at different age. Concerning that the whole sera reactivity is a kind of network-effect which are influenced by many factors, and purified Ig can show the information of the B cell clone that produce 1g. different reactivity of AK auto Abs in the sera before and after purification were compared, and some biological characters of AK auto Abs were analyzed and speculated. On the other hand, based on the previous findings that AK auto Abs are closely correlated with psoriasis, we choose psoriasis patients to analyze the changes of the titer and reactive patterns of AK auto Abs in skin disease, trying to understand the role of AK auto Abs in pathological conditions. -8- 7 AK auto Abs are a group of antibodies that can identify a series of keratin with different molecular weight.
