| The inhibition of melatonin(MT) on growth of human prostatic carcinoma cell lines of PC-3m was studied by examining the influence of MT in a different concentrations and various time on cell culture in vitro with MTT and board clone assays. It was found that MT could inhibit PC-3m cell proliferation under different concentrations and time, its IC50 of MTT and board clone was 41975 u mP?and 435.88 i.~ g mr?respectively. The different concentration of MT and 5-fluorouracil (5-Eu) was simultaneously cultured with PC-3m cell in vitro, the results of absorption degree of MTT and board clone count were investigated and the inhibitory rate were calculated. According to the Jing抯 format , MT could increase 5-Eu to inhibiting PC-3m cell proliferation when O.5? i.~ g mP?of 5-Eu applied with 15.63挆?25 u g ?mV?of MT respectively . The different concentratiotis and time of MT also had effect on PC-3m cell morphogic change ,such as cytoplasm condensed, cell body becoming smaller in microscope, mitrocbondroin swelling in supperrnicrostructure, and it was found that PC-3m cell cycle was changed ,G11G0 was lower G?/M and S phase were higher, and we have found the typical apoptosis under transmission electron microscope(TEM). Our results indicate that MT could inhibit PC-3m cell proliferation, and strengthen or ally to the chemotherapeutic action of 5-Eu, so it maybe decrease the chemotherapeutic dose and adverse drug reaction of 5-Eu in clinical. |
