The Study of the Differentiation-inducing Effect of Tachypksin in Combination with n-Sodium But~ate , GinsenosideRgl on Human Gastric Adenocareinoma BGC-823 CeflsABSTRACTIn this sy, Tachyplesin, a low molecular weight peptide isolated from acid extracts of horseshoe crab (Tachypleus tridentatus) hemocyte debiis, n-sodium butyrate and g~nsenosideRgl were used indi'.idually and in combination with each other to induce the human gastric adenocarcinoma BGC-823 cells~ so that we can compare to their hiologcal effects and investigate the effectiveness of combination treatment on gastric adenocarcinoma cell.The experiment results revealed that the proliferation of BGC-823 cells was inhibited and the cells were arrested in GO/U 1 phase after 2.0 ii g/ml tachyplesin, 2.Ommol/l n-sodium b~Tate or 50 i-i g/ml ginsenosideRgl. The effects of tachyplesin and n-sodiwn butyrate were stronger ~itile that of ginsenosideRgl was weaker. When the BGC-823 cells were treated by 1.0 ii g/ml tachyplesin +1 .Ommol'l n-sodium b~ate or by 1.5 ~ g4nl tachyplesin+50 ii g/ml ginsenosideRgl, their proliferation and cell cycle were also inhibited, the effects were signfficant than those of single treatmentOptical microscope, scanning electron microscope and transmission electron microscope observation showed that the BGC-823 cells which were treated with tachyplesin., n-sodium butyrate and ginsenosideRgl had undergone restomtional morphology and ultrastnrcture changes similar to those of normal cells as follow the volume of cell increased the shape of nuclear became regular, nucleo-cytoplasmic miio decreased, the number and volume of nucleolus decreased ,heterochrumatin reduced while euchroniatin increased, the number and volume of mitochondiion increased and its stmcture appeared wnsistent~, golgi complex turned to be typical , cndoplasmic reticulum and polyribosome increased, microvilli and filopodia reduced, while sheet pseudopodia increased. Tachyplesin inductive effects similar to n-sodium butymie and stronger than ginsenosideRgl, the combination treatment was stronger than the individual treatmentFurthermore, Cytochemical observation showed that the reactive products of AKPasewere decreased. The immunocyiochemical assay revealed that the expression proteins of oncogene C-erbB-2, c-myc, mutant p53 were do~regulated but the protein products of tumor suppressor gene p16 were increased after the treatment of tachyplesin in combinationwith n-sodium butyrate or ginsenosideRgl. And the combination treatment was stronger than the individual treatmentAll of these results showed that the low concentraction of tachyplesin, n-sodium butymte and ginsenosideRgl could effectively inhibit the proliferation of the human gastric adenocarcinoma BGC-823 cells, cause GO/U I cell cycle arrested, change their malignant morphology and metabolism character They could also regulate and interfere the expression of oncogene and tumor suppressor gene con-elative with gastric cancer. In conclusion, tachyplesin has the similar induced differentiation effect to n-sodium butyrate and ginsenosideRgl on the human gasUic adenocarcinoma BGC-823 cells. Furthermore, tachyplesin in combination with n-sodium butymte or gjnsenosideRgl are superior to the individual treatment...
