Pigmented nevus is a common benign skin neoplasm with occasionally recurrent and malignated. Some people found that there are basement membranes around nevus nests and nevus cells. LEA studied on nevus from ultrastructure and immonoelectron microscope level in 80's. He believed that nevus is a kind of intraepidermal structure, as the basement membrane around nevus is immerged into epidermal-dermal junction examinated from serial section. In order to further invested that observation, we reconstructed the 3-D structure of nevus on computer with computer technique and immunohistochemical technique . Aims:l.To study the relationship between basement membrane and nevus cells in pigmented neoplasm.2.To provide a new method for teaching and therapy on nevus.3.To provide a new thought to study on morphology of nevus. Methods:Taken from 10 paraffin embedded tissues, 4 pigmented nevus were cut into serial histologic sections. There are 6 cases of Malignant melanoma, compound nevus and recurrent nevus altogether with cutting normal histologic sections. The expression of nevus cell and basement membrane were respectively applied by S-100 and IV collagen antibody with LAB-SA immunohistochemical technique. Information were inputted into computer after the processes of spelling and recognizing. By the technique of reconstruction, the 3D stereo pigmented nevus models were retrieved from 4 cases pigmented nevus's 2Ddigital data. We observed the stereo structure and clarified and compare the spacial relation between nevus and basement membrane in different pigmented neoplasm. Results:31 Hl.PN comprising several components can be analyzed as a whole or each structure may be viewed seperately.2.It shows the relation between the nevus cell and basement membrane in intradermal nevus.3. It also shows the relation between the nevus cell and basement membrane in junctional nevus.Conclusion:l.the method of the microcomputer aided 3D reconstruction wassuccessfully established in PN.2.The reconstructed 3D image shows PN realistically.3.All structures reconstructed could be displayed alone, in anygroup or totally and rotated in 3D space or rotated continuously atdifferent speed.4. It futher invested the facticity of theory of CLARK, and make a good academic base for clinical therapy to PN.5. To primarily discuss the recurrent causes of PN.6. It can be applied in computer-assisted teaching on dermatology.
