Objective To study the expression andlocalization of N-methyl-D-aspartate receptor(NMDAR) subunits NRland NR2A in the rat retina and the model of rat retina ischemia/reflution respectively . Methods Eighteen Westai rats, feeding in routine way, were randomly classified into six groups. Eighteen right eyes were performed the surgery of ligation of the optic nerve for 60 minutes, and the eighteen left eyes were taken as normal control group without surgery. The rats were sarcrified immediately, 3 hours, 12 hours, 24 hours, 72 hours and 148 hours after 60 minutes of ischemia. The rat eyes was nucleated and sectioned into 10-12um thickness slices on cryostat .We observed the localizations and the changes of NR1 and NR2 in the different groups by immucytochemistry experiment. The results were disposed by image analysis and statistic methods. Results l.The subunits NR1 and NR2A localized in the inner retina, such as ganglion cell layers, inner plexiform layers and inner nucleus layers. The NR1 also'1 ocalized in outer plexiform layers.2.Between control groups and experiment groups, and among experiment groups, the expression of NR1 is no difference, but the NR2A is statistic different. Conclusions 1. the model of rat retina ischemia/reflution was successful. 2.NMDA receptor subunits NR1 and NR2A localized in the inner retina.-2-3. NRl have no relation with the exciting toxic function of glutamate, but the NR2A is related to it. |
