OBJECTTVE: Four different susceptibility methods ofpyrazinamide , Which including acid Ix>wenstein-Jensen'smethods^Cuang's agar medium method, Pyrazinamidase assay and PnCA gene deterrnination, are compared to find out the most suitble one for mycobacterium tuberculosis. METHOD: Sputum samples were collected form 100 smear-positive patients, 100 clinical isolates tuberculosis were got form our laboratory. The susceptibility of ali samples was assessed by acid Lowenstein-Jensen's methods, Kuang's agar medium method, Pyrazinamidase assay and PnCA gene determination. The median time and correlation for final susceptibility results among four methods were compared. RESULT: The correlation betvveen acid Lowenstein-Jensen's methods and Kuang's agar medium method was 99% , Pyrazinamiclase assay was 71%, PnCA gene determinationwas 75%; The median time of acid Lovvenstein-Jensen's methods was 38 days, Kuang's agar medium method was 21 days, Pyrazinamidase assay was 13 days, PnCAgene determination was 4 days.CONCLUSION: Compared to acid Lowenstein-Jensen's methods , Kuang's agar medium method , Pyrazinamidase assay are reliable and rapid, They provide altematives which can adapt to different clinical demand. PnCA gene determination can't be used for susceptible isolates' determination , It has limited worth for clinical vvorko...
