Objective: To investigate the inhibitory ability on hepatitis B virus (HBV) replication and mechanisms of arsenic lrioxide(As2O3), thymic factor D(TFD) and Interferon a-2b (IFNu.2b) in vitro.Methods: ELISA was used to observe the effect of varied concentrations of AsaOa and TFD on the HBsAg and HBeAg in the HepG2.2.2.15 cell line, and the inhibitory rate was calculated. MTT method was used to evaluate the cytotoxicity, and the cell viability and therapeutic index (TI) was calculated. DNA dot blot hybridization technique was also used to compare the amounts of HBV DNA among the AsaOa, TFD treated and untreated cells. At the same time, we took IFNu.2b as a control to evaluate the effect on anti-HBV in vitro. Results: AsaOj, TFD and IFNa.2b inhibited the secretion of HBsAg and HBeAg withrelation to the doses and duration. The results of the dot blotting showed AS2Oa and lFNa.2b inhibited HBV DNA synthesis in a dose-dependent manner but TFD showed no obvious effect on HBV DNA synthesis.Conclusions: As2C>3, TFD and IFNu-2b showed remarkably inhibitory effect on HBV viral protein expression. The results of the dot blotting suggested lhat As20s probably affected HBV DNA synthesis and TFD was likely to affect HBV multiplication in other stages of replication such as viral RNA synthesis and /or viral protein synthesis.
