Objective Previous research shows that 67LR and MMP-7 are important proteins involved in the interaction between cell and extracell matrix, both of which play a critical role in growth, progression and metastasis of malignant tumor. In this study, we want to investigate the relationship between their expression and normal endometrium, endometrial hyperplasia ,and clinical-pathological parameters of endometrial carcinoma, to establish the FQ-RT-PCR method to measure the expression of 67LR mRNA in cells and tissues and explore its correlation with endometrial lesions, to find out the relationship of 67LR protein with its mRNA and make out their clinical values respectively.Methods 52 specimens of endometrial carcinoma, 16 of endometrial hyperplasia ,48 of normal endometrium were evaluated by IHC technique with antibodies against 67LR and MMP-7. The FQ-RT-PCR method for detecting the expression of 67LR was established successfully . 69 specimens of endometrium were detected in the PE Applied Biosystems 5700 Sequence Detector . Results 67LR and MMP-7 showed notably increased expression in endometrial carcinoma and hyperplasia .compared with normal endometrium (PO.001). while there is no significant difference in the former two groups (P>0.05). The expression of the two indicators in carcinoma of Grade 1~2 is strikingly higher than those in Grade 3 (P<0.05), and 67 LR is positively correlated with MMP-7(P<0.05). There is no significant difference in the level of 67LR mRNA among the three groups (P>0.05). The level of 67LR mRNA has no correlation with the 67LR protein(P>0.05).Conclusions The expression of 67LR and MMP-7 is elevated in the early stage development of endometrial carcinoma. Detecting both of them will assist clinical doctors in judging the biological behavior of tumor and directing therapy. Detection of 67LR mRNA may not help diagnose endometrial carcinoma and precancerous lesions.
