| Objectives The goal of this study was to investigate the expression rate of pi 6 p21 andp53 in giant cell tumor of bone ,to study the relationships between any two of the three fators and to study the features of cytology.oncogenesis ,and biological behavior of OCT.Methods. Forty-nine paraffin embedded specimens were examined by immunohistochemical method. 49 histppathologic specimens of giant cell tumor of bone were selected at random,with each cut into 3 section preparations. All section preparations were divided into 3 identical groups ,with each group containing each one of the 49 section preparations. The 3 groups of sections were immunohistochemically detected with pi6 p21 and p53 kits respectively. 2Xk contingency table rank correlation was used to analyse the results.Results (1) The positive expressin of all three genes,p!6 p21 and p53 was observed in giant cell tumor and the positive rates were 71.4%, 85.7% ,38.8% respectively.The positive cells were scattering or massive in distribution. (2)The positive expression of all three genes were seen in giant cell tumor with different histological grades. There was no remarkable significance among the three histological grades in pi6 ,but there was remarkable significance between histological grade III and I ,11 in p21 and p53(p<0.01). (3) The correlation coefficient between pi6 and p21 had high statistical significance ( x 2 =7.2 ,p<0.01),and those between p!6 and p53, p21 and p53 were not statistically significant ( x2 =2.728 and 3.593 ,p>0.05).Conclusions (1) The stromal cells were in a state of abnormal proliferate activites and tumor cells in GCT were those of stromal cells with positive stains of p53 mutationaltype .(2) The expression of p!6 and p21 in giant cell tumor of bone were closely correlated ,while that of p53 was irrelevated to p!6 and p21 .(3) The study suggested that Jaffes classification reflects the malignancy of GCT to some extent ,but has its disadvantage.Shen Wen-rui (orthpaedics) Directed by.
