The Expression Of P-selectin On Platelets And Soluable P-selectin In Serum In Patients With Acute Ischemia

Purpose: P selectin which expressed on active platelets and endothelium is one of the adhesion molecules in selectin family . P selectin is the specific molecule marker of active platelets and endolhlium .At the same time. P selectin is involved in the early stage of the ischemia-reperfusion injury .This primary study is about to investigate the expression of P selectin in patients with acute ischemia and the rc.le it plays in the process of ischemic disease.Methods: 61 cases with acute brain ischemia admitted in hospital were sludied .The paiienls include 20 cases with T1A ;and 41 cases with acute brain infarction ,Their age was from 34 to 88 years old .All the patients were diagnosed by CT or MRI. Diabetes, myocardio infarction, inflammatory, tumor, hepatic disease and renal disease were excluded. 30 persons were selected as normal control simutaneously. The statistical test of the general conditions of the groups w as performed There was no significant difference between the groups (P>0.05). Blood sample were gotten within 48 hours after the onset. 20 of the patients with brain infarction were followed 7 days later. The platelet specimen were centrifuged by 800 cpmicircles per minute) for 10 minute, stored in 4degree centigrade. The serum specimen were centrifuged by 2500 cpm for 20 minute stored in -20 degree centigrade. P selectin on platelets, soluable P selectin in serum, TXB2, 6-keto- PGF1 were measured by ELISA.Results: Compared with the patients with TIA and control subjects, both P selectin on platelets and P selectin in serum were increased significantly (P<0.05), but there was no significant difference between TIA group and control group. In 20 patients followed 7 days later, P selectin on platelets and soluable P selectin were decreased significantly (P<0.05).In patients with acute brain infarction ,a significant correlation was deteced between P selectin on platelets and soluable P selectin in serum(r=0.7765P<0.01) and between both of them and TXB2.No correlation was found between P selectine and 6-keto-PGFl a .There was a significant correlation between soluable P selectin and the volume of infarction.Conclusions: 1 P selectin on platelets is specific molecule marker of active platelets. 2 There is a markable platelet activation in patients with acute brain ischemia, which is the base of the application of anti-platelet medicine. 3 P selectin was involved in the ischemic injury .Maybe blocking the P selectin's function is a new therapy for acute brain ischemia.
