Objective:To detect the expression of C-IAP2 and gas 1 in HL and ALCL at the level of protein and to investigate the roles of the two genes in the mechanism of pathogenesis of HL and ALCL.Methods:HE staining, the monoclonal and polyclonal antibodies CD30(BerH2), CD15(C3D-1), CD45RO(UCHL1) andCD20(L26) were used for selecting the cases of HL and ALCL from 288 cases of HL and ALCL. The clarified HL and ALCL were finally subjected for immunohistochemical staining to analyze the expression of C-IAP2 and gasl in HL and ALCL.Results:1. 26 cases of HL and 19 cases of ALCL were confirmed by HE staining and irnmunohistochemically by CD30, CD 15, CD20, CD45RO staining.2. The positive rate of C-IAP2 in HL was 25/26(96.1%), including 15 cases were strongly stained -HH-, lOcases were stained ++. In 19 cases of ALCL, only 1 case was stained ++, 5 cases were stained +, the other 13 cases were negative. There presented a statistic difference between HL and ALCL groups(p<0.01) for the staining.3. The positive rate of gasl in ALCL was 18/19(94.7%), including 8 cases were strongly stained +++, 9 cases were stained ++, 1 case was stained +, 1 case was negative. Meanwhile in 25 cases HL. the positive rate of gasl was 18/25, among them 5 cases were stained +++, 13 cases were stained ++ or +, the other 8 cases were negative. There presented a statistic difference between HL and ALCL groups(p<0.01) for the staining.4. 2 cases showed to be a mixed type combined with large tumor cells with the expression of C-IAP2 and gasl minicking HL and relalively smaller tumor cells which expressed the c-IAP and gasl minicking ALCL.Conclusions:1. Our study reported the different expression of C-IAP2 and gasl in protein level in HL and ALCL. High expression of C-IAP2 were mainly seen in HL and the gasl was low or negative expressed in most cases of HL, in contrast to relative high expression of it in most ALCL cases. Moreover, the cytoplastic manner of gasl expression implied a lack of its availability of membranous anchoring. The expressing feature of C-IAP2 and gasl also implied a different mechanism of oncogenesis and the different defects in signal transduction of apoptosis pathway in HL and ALCL.2. In few cases, pathohistological features, combined expressions of some lymphocytic markers, C-IAP2 protein and gasl protein in neoplastic cells showed an overlap and a likely transitional state between HL and ALCL.
