As a kind of scaffold material for tissue engineering cornea, collagen is too soft to support cells in stromal layer, and its biodegrading is too fast. In this study, porous collagen sponge were crosslinked using dehydrothermal (DHT) treatment and/or l-ethyl-3-(3-dimethyl aminopropyl)carbodiimide (EDC). The present paper reported detail information about crosslinking such as react conditions and their effect on properties of collagen. The date showed that the mechanical properties of the material were optimized by all the three kinds of treatment, and EDC treatment had greater effect on these properties of collagen. The maximum strength were observed for DHT crosslinking at 120 癈 for 3d. The effect of DHT/EDC treatment on properties of collagen was between that of DHT and EDC treatment. Cell cultivation, pyrogen and hypersusceptibility tests were applied in the biographic evaluation and the results indicated that all the materials were negative and the cell could grow well on the surface of collagen sponge. So the material prepared in this study can be used as the scaffold of tissue engineering cornea.
