P-R andIGFl-s in human duing ear1y pregnancy, we measured P in human serumand studied the variance of P--R and IGFl-3 in human luminal andg1andular ePithelium, endometrial stromal ceII,decidua and tTophoblast cell.lMethods and materialsl 24 uterus endometrium were obtained fromhealthy n1ature early pregnant women(gestational agef5-7 weeks)whorequested fOr abortion, and 6 during mid-secretive phase from thedepallment of patho1ogy(controls). The serum P was measured byRadioimmunoassay; The tissular P--R and IGFBP1--.3 were detected byimmunohi sto chemi stry u sing p o l yc l o n al antib o dy and qu ami fi ed bycomputer image analysis system.The grey scal of positive reaction(GSPR)indicated the intense of positive reaction.[resultl Serum P during the mid-secretive phase gradua1ly reached peakedlevel between gestationa1 5-6 weeks,which was significantly higher thanthat during the mid- secretive phase (P<0.0l), then dectreased to the level atearly gestationa1 5 week. P--Rexpressed in lun1ina1 and decidua,$#R$$$##4KtuI$#&A&A%4mghnffiRtecells,whose variation of exPression showed significam correlations withserum P(r = 0.579, P < 0.0l; r = ~0.773, P < 0.01). P-R also exPressed inglandular ePithelium and troPhoblast cel1s, but had no correlations withserum P.IGFBP-l,IGFBP-2 and IGFBP-3 all exPressed in luminal andglandular ePithelium, and decidual cells. The variation ofIGFBP-l,IGFBP-2 alld IGFBP-3 exPression in endometrium showedsignificant correlations With serum P(rBP-l > 0'44l,pBP-l < 0'0l;rBP-2 30'508,pBP-z < 0'0l; rBP-s>0'373;pBP-s < 0'0l).IGFBPl-3 also exPressed introphoblast cells, but showed no significat correlations with serum P.IConcIusion1 P has an boortan role in early pregnancy: P can stimulatesthe strOmal cell to proliboion and make preParation for decidulization; Palso inhibit the mitosis ofluminal and glandual apithelium and induce glandin endometrium to produce nutrition needed by embryos; P can induce theexPression of cytokine to promote the proliferation of endometrium andtrOphoblast cell. P exed its effect by regUlating the exPression of P-R indiffereflt cells.The exPression of IGFBP-l in endometrium is induced by P, IGFBP-lis an imPortant mediator for P, it may has some roles in decidualization byinteracting with IGF-1; IGFBP-l also associated with the invasion oftroPhoblast cells by interacting with IGF-2; IGFBP-l can enhance thecombination of IGF-l,IGF-2 and IGF-R by dePhosphorization, so toregulate its effect on the developmam of tissues.P can stimulate the expression of IGFBP-2 and IGFBP-3.They are4important mediators for P and may have some roles in decidualization; Besides, IGFBP-2 and IGFBP-3 may regulate the effect of IGF-2 on embryos by interacting with IGF-2, and they may also regulate the effect of IGF-1 in early pregnancy.
