The drinking of potions of decoctions of the leaf of the guava plant,Psidium guajava L.(Myrtaceae),as well as chewing of the fresh leaf,had been employed in the treatment of acute diarrhoeal diseases in folk medicine,especially in tropical areas of the globe,such as Brazil,Guangdong and guangxi in China.This study's objective is:(1).To explore the leaf of the Psidium guajava and its extract effect on rotavirus in vitro.(2).to study the leaf of the Psidium guajava effect on rotavirus in vivo.By this way to support the clinic treat enteronitis caused by HRV in theory,and to exploit new drugs grade II of country basied in it.Methods (1).Essential oil, quercetin and ursolic acid were extracted from the leaf of Psidium guajava,and we studied three ingredients effected on rotavirus in vitro;(2).Lay out normal control group ,model group of infected rotavirus,treatment control group and treatment group of the leaf of the Psidium guajava,model was used by gastric gavage with rotavirus, observe the ratio of negative of antigen-rotavirus from stool ,and the content of SIgA from stool by RIA method ,and the pathological changes of small intestines from mice by HE method, studied the leaf of Psidium guajava effected on the mice infected with rotavirus.Results (1).The cell MA-104 grow well before the cell were foreclosed by the sera of essential oil, quercetin ,ursolic acid and the leaf of Psidium guajava and then infected by rotavirus; The cell MA-104 grow well before the cell were infected by rotavirus and then joined the sera of Essential oil, quercetin ,ursolic acid and the leaf of Psidium guajava; The cell MA-104 grow well before the cell were infected by rotavirus which were mixed with the sera of essential oil, quercetin ,ursolic acid and the leaf of Psidium guajava;the results were obviously preceded to control group of rotavirus which detected by ELISA(P<0.01). (2).48 hours late,the mice appeared diarrhea,took little food et al which infected by gastric gavage withrotavirus;72 hours late they appeared dehydrate,skin loosen,diill and motionless;4days late they appeared symptom of failure and 5 days late someone dead,which were most obviously and seriously is control group of infected,the treatment group of great dose is well.The amount of dead mice,the amount of positive which detected by ELISA from stool,the content of sugar and natrium from stool,which the group of the leaf of Psidium guajava is under the control group of infected and the control group of treatment.The content of SIgA from stool which is the group of the leaf of Psidium guajava surpe to others.The slice of pathology showed: the control group of infected the small intestines mucosa grew thiner,mucosa grew swelling and congesting,many lymphocytes infiltrated,villus failed off;The small intestines mucosa didn't swell and congest in the group of the leaf of Psidium guajava,little lymphocytes infiltrated under the mucosa and the villus were well; The small intestines mucosa grew swelling and congesting in the control group of treatment,many lymphocytes infiltrated under the villus.Conclusion The experiment in vivo and in vitro were studied in this article.The conclusion were as follows: (1).the leaf of the Psidium guajava and its extracts essential oil, quercetin ,ursolic acid can reduce the virulence of rotavirus,lower the infection of rotavirus,swell the resistance of MA -104 to rotavirus; (2).The leaf of the Psidium guajava and its extract can alleviate diarrhea of the infected mice,extent of dehydrate and lower the ratio of death of the infected mice.They can reduce the antigen-rotavirus of stool from mice,accelerate the small intestines absorbing sugar , natrium and excreting SIgA.They have protection to the small intestines...
