The multi-drug resistance is an important factor to induce the failure of chemotherapy in human cancers. Through experimental method design and clinical application, this paper investigated the detecting method and clinical significance of multi-drug resistance gene (MDR-1) expression. The detecting method of multi-drug resistance gene (MDR-1) expression was established by the reverse transcription polymerase chain reaction (RT-PCR). Because of its efficient, sestive and economic, the detecting method has wide clinic application. The experimental results and analysis showed that MDR-1 is the objective mark on detection of the samples. The MDR-1 displayed high expression at the different degrees in some cancers.Used human multi-drug resistance KVB200 cell line as the cell model, the reversal effects of a Chinese medicine reversal agent, multi-drug resistance gene antonym chain, cell differentiation agent and hyperthermia were investigated by the experiments. The paper explored the relationships among multi Chinese medicines, antonym technology, hyperthermia and cell differentiation agent to the reversing of multi-drug resistance and inducing apoptosis in cancer cell. The experimental results expressed: that TTD, RTD, CDA-II and hyperthermia were obviors on reversal action; LTZ and MDR-1 antonym RNA were some reversal action; there was cooperative action in combination of TTD with MDR-1 antonym RNA, CDA-II and hyperthermia. CDA-II and hyperthermia could induce KVB200 to apoptosis.Through the MDR-1 expression, observation and visitation of chemotherapeutic effects and survival rate of ovarian carcinoma in 53 cases, investigated the position of the MDR-1 expression on clinical application. The results indicated obvious relationship between the negative or positive expression of MDR-1 and chemotherapeutic effect. The chemotherapy could induce the drug resistance to occur, or that MDR-1. On the clinic, MDR-1 detection and reversal method research could decrease the resistance to occur and guide the use of chemotherapeutic drug. The clinical observation on ovarian cancers discovered that the combining use of pre-operation chemotherapy and CDA-II could decrease the MDR-1 expression, control ascites and reduce tumor dimension. CDA-II may change the methylation state of tumor cell to induce apoptosis, reserve multi-drug resistance and enhance the clinicaleffectiveness.
