study the differences of morphology ,phenotype and function of dendritic cell(DC) derived from the healthy individuals and the patients with nasopharyngeal carcinoma (NPC).Methods DC were isolated and developed from peripheral blood monocytes harvested from 12 NPC patients and 9 healthy volunteers in vitro, and were studied in terms of shape ;we also detected the percents of DC expressing phenotype of CD83,CD1a,CD86 , HLA-DR and their fluorescence intensity by flow cytometry ;then, MTT method was uesed to measured the capability of DC in mixed lymphocyte reactions(MLR).Results The shape of DC derived from monocyte in the healthy volunteers' peripheral blood was more typical than that of NPC patients';and the expression rate of CD83,CD1a on DC of healthy volunteer's group(24.3±14.3%, 40.5±22.6%) was significantly higher than that of NPC patient's group(8.6±10.7 %,15.0±10.6%)( P<0.05); the healthy volunteer's DC had the higher expression level (4.80±3.27 and3.68±1.96)of CD1a and CD83 than that of NPC patients'(3.26±1.49,2.70±0.74),but not significantly(P>0.05); DC of healthy volunteer's group could trigger proliferation of allogenous lymphocyte(P<0.05)while that from NPC patient's group could not significantly(P>0.05);and the stimulating index(SI) of NPC patient's group was lower than that of healthy volunteer', but that also had no statistic difference(P>0.05). Conclusion These results reveal that under the same condiction, the yield of DC derived from both NPC patient's monocyte in peripheral blood is lower than that of healthy individual's,and DC from NPC patient may be in an immature state and could not trigger the proliferation of lymphocyte.
