Objectives: To observe the effects of extraction of Peristrophe roxburghiana on the systolic blood pressure, renal function and hemorheology in the renal hyprtensive rats and the histological changes of heart, kidney. To study the antihypertensive mechanism of HSX briefly too.Methods: (1) Baseline blood pressure (BP) was measured, and modified 2 kidney-1 clip (2K1C) operation was given. (2)Four weeks later, except sham group, hypertensive rats were randomly allocated to five groups such as HSX(i.e herb 3.42 g / kg ), HSX(i.e herb 1.14 g / kg), HSX(i.e herb 0.38 g / kg), Captopril(Cap, 20 mg/kg) and model group. After allocation, drugs were administered by i.g respectively. Rats in model and sham group were administrated with identical volume of distilled water. The treatment period was 7 weeks but discontinued in the 6th week. (3)The blood pressure was measured weekly. At the end of the 7th week, the urine volume, urine protein concentration, blood urea nitrogen (BUN), creatinine (Cr), hemorheology parameters and angiotensin II (Ang II )were measured too. Kidneys and hearts were collected to observe the histological changes.Results: (1) After 1 week of administration, HSX(H) had potent antihypertensive effects(P < 0.05 vs model group) which had lasted for 5 weeks. The BP in HSX(M; L) lowered significantly after 2 weeks of administration(P < 0.05 or P < 0.01 vs model group). After withdrawal in the 6th week, the BP in all groups rose again. When the rats were treated again, the BP in Cap, HSX(H) and HSX(M) lowered significantly once more(P < 0.05 or P < 0.01 vs model group). During the course of administration, Heart rate(HR) didn't change markedly.(2) Compared with those of sham group, the urine protein concentration, total urine protein/24 hour and BUN of the model group increased significantly. HSX(H,M and L) and Cap reduced the total urine protein/24 hour & BUN remarkably. (3) Hb, Hh and RI increased significantly in the model group (vs sham) but decreased significantly in the HSX(M) group (P < 0.05or P <0.01). Hb (10s-1) decreased in the HSX(H) group too (vs model group, P < 0.05).(4) Ang II values in hearts and kidneys of model rats increased significantly in the model group (vs sham) but decreased significantly in the HSX(H,M,L) and Cap groups (P < 0.05or P < 0.01). (5)The myocardial collagen and glomerular pathological injuries were markedly prevented in HSX(H) and Cap group.Conclusions: HSX can reduce the blood pressure slowly and mildly, which is in part related to RAS, and has no effect on the HR. It can exert the renoprotective effects by lowering total urine protein (24h), BUN. It can lower hemorheology parameters, too.
