| Objective: To study the effect of JinShuiLiuJunJian on histomorphological and stereological changes of cilia and ciliated cells of tracheal epithelium and bronchiolar epithelium in mice with chronic bronchitis, and make research on the internal mechanism of treating experimental chronic bronchitis with JinShuiLiuJunJian. Methods: The male NIH mice were randomly divided into normal group, model group and JinShuiLiuJunJian treated group, and each group consisted of 10 animals. We duplicated the animal model of chronic bronchitis with refined smoking-fumigated method and made intervention with JinShuiLiuJunJian oral liquid. Scanning electron microscope (SEM), light microscope and transmission electron microscope (TEM) methods, image analysis and morphometric quantification were used to observe and measure the ultrastructure histomorphological and stereological changes of the cilia and ciliated cells of tracheal epithelium and bronchiolar epithelium in the mice of the above 3 groups after the mice of JinShuiLiuJunJian treated group were fed for 4 weeks. Results: The numerical density on area of the ciliated cells of main bronchiolar epithelium, the regularity of the cilia of tracheal epithelium and the numerical density of the cilia of bronchiolar epithelium of the model group were significantly reduced than the normal group (P<0.01), while the diameter, the proportion of vesiculation and compound cilia and giant cilia of the cilia of bronchiolar epithelium were significantly raised than the normal group (P<0.01). Compared with the model group, the numerical density on area of the ciliated cells of main bronchiolar epithelium, the regularity of the cilia of tracheal epithelium and the numerical density of the cilia of bronchiolar epithelium of JinShuiLiuJunJian treated group were significantly increased (P<0.01 or P<0.05), while the diameter, the proportion of vesiculation and compound cilia and giant cilia of the cilia of bronchiolar epithelium were significantly reduced (P<0.01 or P<0.05). Conclusions: In mice with chronic bronchitis, the cilia of tracheal epithelium and bronchiolar epithelium weredecreased, thickened, curved, detached, and adhered to each other, and the percentage of axonemal ultrastructure abnormalities (AUA) of the cilia were increased, and the ciliated cells of tracheal epithelium and bronchiolar epithelium were decreased and detached. JinShuiLiuJunJian oral liquid can obviously alleviate above-mentioned pathomorphological changes in mice with chronic bronchitis on a quantized level and treat chronic bronchitis effectively. |
