Subject: To probe into the therapeutic effect and mechanism of ZI ZU HUO XUE BU QI FANG (including salvia miltiorrhiza bunge, ligusium wallichii franch, angelica sinensis (oliv) diels, scutcllania baicalensis georgi and astragalus) on the intrauterine growth retardation (IUGR) according to determination and analysis of serum nitric oxide (NO), plasma endothelin-1 (ET-1) and urine 8-epi-prostaglandin F2α (8-epi-PGF2α).Method: Pregnant rat model with IUGR was established using passive smoking method. Fifty pregnant rats with IUGR were randomly divided into five groups: Group non-intervention, and Group intervention (with high-, middle- and low-dose Chinese traditional medicine, and L-arginine). In addition, ten normal pregnant rats were selected into Group control. Group intervention were administered 3ml/100g·d, lml/100g·d, 0. 3ml/100g·d of ZI ZU HUO XUE BU QI FANG, and 0. 05g/100g·d of L- arginine, respectively. Urine samples from the female rats were collected before and on the 21th day of the pregnancy; Blood samples were collected and their fetuses were taken out from the killed pregnant rats. The weight, nose-hip-length and poundera index of these fetal rats were measured. Serum NO content was determined with nitio-reductase method, plasma ET-1 concentration was assayed by radioimmunoassay (RIA), and urine 8-epi-PGF2α level was determined with enzyme immunoassay (EIA) from the pregnant rats. The change in placental tissue and cell was observed under the microscope.Result: ① Compared with Group intervention, Fetal weight was obviously decreased (p<0. 01), the distance between nose and hip was markly shortened (p<0.01) and the poundera index was apparently lowered in those fetuses from Group non-intervention (p<0. 01). ② In Group control, 8-epi-PGF2α level at the end of pregnancy was obviouslyhigher than that before pregnancy (p<0. 01). ③ Compared with Group intervention, serum NO level was obviously lowered (p<0. 01), plasma ET-1 level was clearly increased (p<0. 01), NO/ET-1 was apparently decreased (p<0. 01) and urine 8-epi-PGF2α, level was increased (p<0. 01) in Group non-intervention. ④ At the end of pregnancy, urine 8-epi-PGF2α levels in Group intervention with different Chinese traditional medicine had not diffence (p>0. 05) , but were obviously lower than that in Group intervention with L-arginine (p<0. 01) . ⑤ Pathomorphological change in placental tissue and cell, such as lymphocyte infiltration in the decidual zone, and decreased blood sinus and proliferated deutoplasmic cell in the labyrinth zone, happened in Group non-intervention.Conclusion: ① There is a lipid peroxidation in the normal pregnant rats. ② There a markly increased lipid peroxidation and a NO/ET-1 ratio imbalance in IUGR pregnant rats. ③ ZI ZU HUO XUE BU QI FANG has good therapeutic effect on IUGR since it can give play to an antioxidation function, inhibit increased lipid peroxidation and regulate NO/ET-1 balance.
