IntroductionSinusoid capillarization is an important pathological change during the formation of liver fibrosis (liver cirrhosis) , and the formation of the sinusoidal subendothelial basement membrane is a cardinal feature of it. At present, there are few reports on " the process of sinusoid capillarization" in the formation of liver fibrosis of non - alcohol induced liver disease, so it is remarkably essential to do systematic research on the dynamic changes of laminin ( LN) , which is the main component of basement membrane, in the formation of liver fibrosis. Successfully, we use Buyssens methods to set up the hyperchoester-emia rabbit, which accompany with the fatty liver and liver fibrosis model. Immunohistochemistry methods and transmission electron microscope ( TEM) were used to outline the perisinusoidal changes in rabbit s liver fibrosis, in order to find out the relationships between sinusoid capillarization and liver fibrosis.Materials and Methods1. Grouping Division and Model ConstructionForty male New Zealand White Rabbits were randomly divided into 2 groups, that is, fifteen in normal control group and twenty - five in model group. The rabbits in model group were given high steroneanimal feed, i. e. adding 2% cholesterol on the common animal feed. They were administrated once a day in the morning for 8 weeks, while the control group was given common animal feed for 16 weeks. Rabbits were killed at the end of 2 weeks, 4 weeks, 8 weeks, 12 weeks and 16 weeks, i. e. model group 5 rabbits each time and control group 3 each time, respectively.2. The Pathological Detection of Hepatic TissueAfter the celiac anaesthetizing and fixation by heart perfusion of rabbits, cut the hepatic tissue from the central part of right lobe of liver. Fix it with 40mg ?L'1 formaldehyde solution. HE and VG - picric acid stains were used to evaluate the liver histopathological alterations and the degree of steatosis and fibrosis. Dye LN by immunohistochem-ical technique to observe the dynamic changes of sinusoindal wall in the process of liver fibrosis. Use the pathological image analysis e-quipment to analyze the result, and then measure the percentage of the area of collagen and LN. Take another hepatic tissue, fix it by 25mg ?L"1 glutaraldehyde solution, and observe the ultrastructure morph changes of it by TEM.3. The Determination of Hyaluronic acid ( HA)Use radioimmune method to observe the endothelial cell injury of hepatic sinus.Results1. The Histopathological Changes of Hepatic Tissue1. 1 The Light Microscope (L. M. )HE stains: In model group, slight steatosis and spotty hepatocel-lular necrosis were observed after 2 weeks. At the end of 4 weeks, we cant observe the aggravated steatosis of hepatic cell, but the inflam-matory infiltration of cells aggravate. At the end of 8 weeks, more cells become necrosises, and inflammatory cell infiltrate into conglo-bation. After stopping high sterone feed, steatosis, as well as, necrosis and inflammation abate.VG - picric acid stain; At the end of 2 weeks, collagen increased gradually. At the end of 8 weeks, terminal vein (including central vein ) thickened, the collagen extend to sinusoid, and perisinusiodal fibrous tissue increased. After stopping high sterone feed, collagen decreased gradually, but is still higher than normal, even until the end of 16 weeks. Pathological image analysis showed the rates of collage in model group were obvious higher than that in control group (p <0.001).1.2 Transmission Electron Microscope (T. E. M. )In model group, at the end of 2 weeks, parts of the cytoplasm' s distal fenestra number of sinusoidal endothelial cells ( SEC ) decreased , and there is no formation of basement membrane in subendo-thelium. At the end of 4 weeks, the distal fenestra number of SEC near the septum decreased or disappeared, and there were imperfect basement membrane. At the end of 8 weeks, the distal fenestra number of SEC near the fibrous septum decreased or disappeared, and there were complete basement membrane und...
