IntroductionOsteosarcoma is the primary malignant tumor of bone and the first position in them , It is highly malignant and easy to occur transferring into lung in early stage , prognostication is worse . Giant cell tumor of bone is commoner and the incidence is only minor to osteochondroma in china , belong to potential malignant or between benignant and malignant tumor that can dissolve bone , the recurrence rate is high after curettement . There are no effective items for diagnosis , pathological grades , therapy and prognosis judgement of these two bone tumors till known .p19( ARF) is tumor suppressor gene that was discoveried recently and one of the INK4 ( cell cycle dependent kinase inhibitors ) family members . Its gene location superposes with that of p16 (INK4) and together are called INK4/ARF location. p19 protein is nucleus protein , and it widely distributed in the postnatal tissue . The over expression of p19 gene and protein can results in stagnancy in Gl and G2 stage cell cycle , thereby , interdicts the over proliferation of cell and playsan role in inhibiting the grow of tumor cells . Its action mechanism is determined by stabilizing wild p53 . p19 is combined with Mdm2 in nucleolus and interdicts Mdm2' s combination with p53 and the function of transporting into cytoplasm to resolve , so as to boost up the effect of inhibition cancer of wild p53 . p19( ARF) ' s action and status in the grown of tumors is in the stage of pilot study . The experiment aims at discussing the expression in bone tumor , making farther studies .MaterialsAll cases were from Department of Pathology , the First Hospital , China Medical University , and from 1992 -2001 , including 25 os-teosarcoma , 34 giant cell tumor of bone and compared to 23 osteo-chondroma . Giant cell tumor of bone is divided into three groups by Jaffe grading , including 15 grade I , 15 grade II and 4 grade Iff of giant celltumor of bone . All cases have routine HE stain and are re-newedly determined by three pathologist . Agent in the experiment , pi9 monoclonal antibody is from Boster company , S - P , 10% goat serum and second antibody marked by biotin are from Maixing company in Fuzhou .MethodsThe experiment uses the sensitive and differential S - P method to detect the expression of p19 protein in 25 osteosarcoma and 34 giant cell tumor of bone and compared to 23 osteochondroma , analyzes the expression of p19 protein in different tumors of bone , discusses itsrole in occurring and development of bone tumors .ResultsIn 25 osteosarcoma, only 1 nucleus is stained brown , the positive rate of p19 is 4.00% . In 15 grade I giant cell tumors of bone , positive rate of p19 is 26. 67% , In 15 grade H giant cell tumors of bone , positive rate of p19 is 13. 33% , 4 grade Iff cases are not stained , general positive rate is 17.65% . In 23 osteochondroma , 11 cases appear stain in nucleus , positive rate is 47. 83% . x2 test is applied for statistic disposal . Significant differences can be seen between osteosarcoma and osteochondroma , P < 0. 01. Significant differences can be seen between giant cell tumor of bone and osteochondroma , P <0.05 . No differences can be seen between osteosarcoma and giant cell tumor of bone , p >0. 05. No differences can be seen between grade I and II giant cell tumor of bone , No differences can be seen between grade I and H ~ Iff giant cell tumor of bone , Because the cases grade of giant cell tumor of bone is only 4 , the result does not be compared in groups.DiscussionsOsteosarcoma is the common primary malignant tumor of bone and 21% in them, It is highly malignant and easy to occur transferring into lung in early stage, prognostication is worse . Giant cell tumor of bone is commoner and the incidence is only minor to osteochondroma in china, belong to potential malignant or between benignant and malignant tumor that can dissolve bone , the recurrence rate is high aftercurettement, up to 30% ~ 50% , a few cases may transform into malignant or transfer into lung.
