| Hantavirus(HV), which belongs to Genus hantavirus, Family Bunyaviridae, is single-stranded, negative and segmented RNA virus. Its RNA genome consisting of the segments L, M, S, which encode the RNA-dependent polymerase, glycoprotein Gl and G2, and nucleoprotein(NP), respectively. Recent data indicated that there are at least 30 serotypes or genotypes circulating in nature.Hantavirus infection is becoming a globle serious problem in the field of public health with its wide epidemic range and the severity of the diseases. As we know, two main serious diseases are caused by the virus, which are Hemorrhagic Fever with Renal Syndrome(HFRS)(or EHF)and Hantavirus Pulmonary Syndrome(HPS). It is most serious in China with more than 90%( 150,000-200,000) of the worldwide .Direct and Indirect Immunoflorescent Assay(IFA), nest RT-PCR and nucleoacid sequencing of type-specific partial M segment(1984~2317nt) were used in this study to samples in JiLin province and ShenYang city in China and virus isolation was used by cell cuture. From lung tissues of two Apodemus peninsulae captured in JiLin province, two hantavirus (JI89 and JI93) were established in cell cuture, and whole small(S) genome segments of isolate(JI93) was sequenced. RT-PCR and Phylogenetic analysis indicate that JI93 is a new subgenotype of HTN , and its host rodents are Apdemus peninsulaes.The sequences from 20 positive samples were analyzed by alignment and phylogenetic tree analysis. One subgenotype(JP4 JiLin province) of SEO and one subgenotype (Shen43 ShenYang city) of SEO are found by comparison of nucleotide sequences .And then combined with the epidemiological data, the kinds of rodents earring the virus were identified and the distribution of genotypes and subgenotypes in JiLin province and Shenyang city in China was undertaken. The large-scale molecular epidemiological study was took into action. All of these would be provide a solid basis to use more effective preventive methods in different foci in prevention and treatment of the disease. |
