Objective: Frozen heterotopic rat bone allograft was studied to evaluate the role of histocompatibility on bone allograft acceptability.Materials and Methods: Groups of rats, syngeneic or differing with respect to major histocompatibility loci, were studied. Grafts were studied histologically at 5,7,14,28 and 42 days after transplantation. In addition, the state of immunity was evaluated in vitro by immunohistochemistry to determine the cell-mediated responses and ELISA to determine the serum-mediated responses.Results: Histologically, different immune rejection was noted. In vitro analyses failed to reveal, if any, either antibody or cell-mediated responses in syngeneic rats after bone transplantation. In contrast, rats with grafted bone from donors that had major differences responded strongly.Conclusion: Histocompatibility seems to play a very important role in rat bone allograft transplantation. The immune response in the rat seems to influence the histological fate of frozen bone.
