Assessment Of Interferon-r And Interleukin-10 In Studying Th1/Th2 Subset Swift Of Patients With Renal Cell Carcinoma

Objectives To study Thl/Th2 subset swift and anti-tumor immunity capacity of patients RCC, create theory basis for early diagnosis, prognosis judgement and immunotherapy of RCC.Methods 40 patients with RCC and 20 controls were studied, the levels of interferon-r and interleukin-10 were investigated by ELISA in peripheral blood of all , renal vein blood of 11 patients during operation and peripheral blood of 12 patients about 1 month after radical nephrectomy .Results (1 ).IFN-r level(74.62±9.43pg/ml)in peripheral blood of patients with RCC was significantly lower (p<0.01) than that(96.88±40.27pg/ml) in controls, however, IL-10 level(95.88±32.87pg/ml) of patients with RCC was apparently higher(p<0.01)than that of controls (71.19±14.10pg/ml). (2). In 11 patients with RCC , IFN-r level of renal vein blood during operation was apparently lower(p<0.01)than that of preoperative peripheral blood, reversely, IL-10 level of renal vein blood was higher (p<0.05) . (3). Compared with that of preoperation , IFN-r level of 12 patients investigated was higher(p<0.05) about 1 month after operation ;IL-10 was significantly lower(p<0.01) than before. (4). IFN-r and IL-10 levels varied according to different stages, IFN-r level decreased with higher stage, while IL-10 level increased.Conclusions Higher IL-10 level and lower IFN-r level of peripheral blood in patients with RCC verified that Th2 cytokines expression was enhanced in patients with RCC, suggesting that there was Th1-Th2 swift in patients with RCC. With tumor progressing, plasm cytokines levels of patients with RCC changed , IFN-r decreased gradually, while IL-10 increased, showing that Th1-Th2 swift in patients with RCCincreased with tumor progressing .The enhanced Th2 cytokines expression of perioperation was improved during operation in RCC patients; however was repressed about 1 month after radical nephrectomy, also suggesting that Th1-Th2 swift in patients with RCC was intensified during operation and was reversed to some extent after nephrectomy. Thl cells dominate cell immunity , playing an very important role in anti-tumor immunity, cytokines (IL-2, IFN-r eg)mainly produced by Thl cells could strengthen the cytotoxic effects of T cells, K cells and NK cells; while Th2 cells lead humoral immunity and suppress cell immunity, cytokines(IL-4, IL-10 eg)produced mainly by Th2 cells could suppress Thl cells to produce cytokines , repressing the cell immunity of body. It's the crux of anti-tumor for body to keep reasonable Thl/Th2 balance, once Th1-Th2 swift happens, the anti-tumor activity would be suppressed, causing tumor's escape from immunity. Inhibiting Th2 cytokines' strong expression and adjusting Thl/Th2 subsets balance will be another important topic relevant to tumor immunotherapy.Postgraduate Wang KeDirected by Dong Shengguo Professor...
