Objective:The purpose of the present study was to search for morphologic evidences for three P -adrenergic receptor(P-AR) subtypes (PI, P2 and P 3) mRNA in cells from lymphoild tissue, to study which P -adrenergic receptor subtypes mRNA positive cells was the most one than the others in lymph node tissue, to provide evidences in order to probe into the mechanisms and paterns of the relation between sympathetic system and immune system. Methods: Choosing the the frozen section of 24 cases of human lymph nodes , 2 cases of lymphoid tissue of small intestine tunice mucosa and 2 cases of lymphoid tissue of spleen,we detected the presence of P 1, P 2 and P3 adrenergic receptors mRNA in the cells of these lymphoid tissue by in situ hybridization (ISH). We count the positive cells per 10 high-power fields (/10HPF) using light microscope, and compare the amount of three P -adrenergic receptors positive cells in 24 cases of lymph node. Results: The distribution of P 2-AR positive cells is not regular except a few positive cells were around the vasculature and lymphatic channels (the region that more sympathetic nerve fibers innervated). The distribution of B1-AR andB3-AR positive cells were not regular. The mean amount of B-AR is: B1-AR(16.54 ±1. 08/10HPF) ; P 2-AR (171. 17 ± 64. 63/10HPF) ; B 3-AR (31. 13 ± 7. 04/10HPF). The amount of P 2-AR positive cells is the lagest one in three B-ARs and showed significant difference (P<0. 01). The amount of P 1-AR and B3-AR showed no significant difference (P>0.05). Conclusion: B-AR mRNA indeed exist in human immune cells and norepinephrine may have direct effects on immune cells through P- ARs on immune cells.
