Aims:1.To study the expression of E-CD/catenin complex, E-CD, β-catenin exon3 gene abnormal in non-small cell lung cancer. 2.To research the expression of E-CDmRNA and its clinical pathological value in non-small cell lung cancer.Methods:1. Using PCR-SSCP and sequencing analysis, we examined abnormality of E-CD and β-cat genes in 24 non-small cell lung cancer cases and two cell lines(A549, SPC-A1). 2. Using RT-PCR, we examined E-CDmRNA expression in these non-small cell lung cancer cases and cell lines.Results: 1. Abnormal band shifts were found by PCR/SSCP at exon9 in 2 of 24 non-small cell lung cancer cases, no band shifts was found at β-catenin exon3. 2. E- CD exon8 band shifts were found in SPC-A1, β-catenin exon3 was found in A549. 3. Sequencing analysis of the abnormal band in one patient revealed T base inversion at codons 50(50+T), and 82(82+T),G base at code89(89+G)respectively ,and that in the other patient showed T→C transition at codon 94,G deletions at codons 171(171-G) and C deletions at codons 190(190-C), respectively. 4. β-catenin exon3 in A549 manifested G→A transition at codon 556,C→G transition at codon 557, i.e, GCT→AGT. 5. RT-PCR revealed deletion or decrease of the E-CD exon8-9 mRNA in 10 NSCLC cases and decrease in both cell lines. 6. There were significant correlation between the abnormal expression E-CD and the lymph node metastasis , the differentiation degree and tissue type. Namely, the abnormal expression of E-CD in lymph node metastasis was higher than non lymph nodemetastasis; squamous carcinoma higher than adenocarcinoma; and low differentiation degree carcinoma higher than high-middle ones (P
