Peripheral nerve injury is one of the tough problems in neuroscience.It is a hot topic that research the factors of promoting nerve regeneration and the changes in the microenvironment of peripheral nerve in recent years. The rats sciatic nerve was transected and then bridge-connected with silicone tube.In the first part of the expriment the images of serial slices were taken by micro-photograph system and inputted into computer by which the image matching and 2-D integrate processing were completed. The 3-D reconstruction and display was accomplished in SGI workstation. We also study the variation of S-100αand laminin by immunohistochemical methods in the period of degeneration and regeneration following sciatic nerve injury. In the second part of the experiment we investigated the effect of local application and target muscle injection of nerve growth factor and ciliary neurotrophic factor on the regeneration and functional recovery of transected peripheral nerve. The methods of sciatic nerve functional index measurement, Electrophysiolgical assessment and morphometric analysis were applied on all the animals.Result 1. Soft red crisp colloid was seen in the silicone tube at 3 and at 7 days after injury, while a soft red strip connecting two sides of transected sciatic nerve was found 15 days later. From 30 to 90 days after injury, the neonatal nerve became thicker ,more tenacious,and its color became brighter than before. 2. Wallerian degeneration was observed in proximal and distal segment oftransected sciatic nerve at 3 days after the injury. Degree of degeneration is more serious in distal part than that in proximal part. The neonatal nerve from proximal part began to step into sillion tube at 15 days after the injury. At the same time, Schwann cells were phagolysising degenerated myelin sheath on the electron microscope section. The distal nerve has degenerated completely, and the neurilemma tube started to form. Myelinated nerve fiber was found in the proximal, middle and distal segment after 30 days. The amount of myelinated nerve fiber become more, and its diameter became thicker at 30 and at 90 days after the injury. Morphometrics of some neonatal nerve was similar to that of the normal nerve. 3. Compared with normal nerve fibre, some difference in the neonatal nerve fibre was showed by the 3-D images: node of Ranvier hadn't formed and its array was turbulent. In addition, both the myelin sheath and the axis-cylinder were thinner, and the neurolemmal tube had formed.4. S-100α is expressed in plasma of Schwann cell and myelin sheath. S-100α started to rise at 3 days after injury. The quantity of S-100α of proximal and distal segment reached peak value at 30 days after injury and was more than that of normal sciatic nerve until at 90 days. At the same time point, The quantity of S-100α of proximal was more than that of distal, and differentia is obvious(p<0.01). S-100α in middle segment started to rise at 15 days after injury, and reach peak value at 90 days after injury.5. Laminin is expressed in matrix and inner basilar membrane of peripheral nerve.Concentration grads of laminin was obvious in the period of regeneration. Laminin started to rise at 3 days after injury. The quantity of laminin of proximal and distal segment reach to peak at 30 days after injury,then started to drop.But the quantity of laminin of proximal was more than that of distal, differentia is obvious(p<0.01).And until 90 days later, The quantity of laminin of proximal and distal segment was more than that ofnormal sciatic nerve. Laminin in middle segment started to rise at 15 days after injury. The quantity of laminin in middle segment was more than that of distal but fewer than that of proximal. Differentia is obvious(p<0.01). The quantity of laminin of middle segment reach peak value at 60 days after injury.The differentia of laminin in proximal, middle and distal segment was not obvious(p>0.05).6.The SFI, electrophysiologic parameters(nerve conduction velocity and...
