Vascular dementia (VD) is an acquired syndrome of intelligence malfunction caused by cerebral vascular diseases including cerebral hemorrhage, ischemia and global cerebral ischemia and hypoxia. The main clinical symptom is the descending of memory and cognitive dysfunction. Some may attend by language barrier, dyskinesia, disorder of spatial resolution and personality changes. The rate of occurrence is differentiated by researched objects, methods of filtration , diagnosis criterion and investigation periods. In Occident, VD account for 10-20% senile dementia, but in Asian it's much higher because of the high rate occurrence of stroke .In China, VD accounts for 60-70% dementia and continuously ascends resulted from increasing of the elderly .VD seriously affects patients living quality, at the same time brings a heavy burden to families and our society. In the recent years, people draw a lot of attention on VD, thus both of fundamental research and clinical investigation are developed.The traditional Chinese medicine is a big treasury. Much research about effects of traditional Chinese medicine on prevention and cure of VD is progressively conducted. Significative achievements are gained no matter decoction or monomer and effective part extracted from a single Chinese herb. Compound 9714, as a medicine that mainly cures VD is a monomer extracted from Anemarrhenasaponin by Institute of Radiation Medicine, Academy of Military Medical Science. Based on pilot study, it is proved to have an effective action on anti-cerebral ischemia, increasing cerebral blood flow, inhibiting platelet aggregation and improving learning and memory ability and so forth in models of VD.Our study continues to explore on the protective effects of compound 9714 on VD models and its mechanisms. The main experimental results are listed as follows:1 the effect of compound 9714 on the ability of learning and memory and free radical injury of model mice induced by D-galactoseD-gal(100mg/kg) hypodermic on back and neck for 42~44 days results in model mice descending of learning and memory ability and free radicals injury. Compound 9714(15mg/kg, 30mg/kg, 60mg/kg) can improve learning and memory ability and alleviate free radicals injury in model mice. In the water maze test,compound 9714 makes model mice shorten arrival time and reduce the frequency of errors (P<0.05,P<0.01,compared with the model). Moreover, we observed that it also can enhance the vitality of SOD (superoxide dismutase), decrease the content of MDA(malondialdehyde) and increase the content of GSH (Glutathione) in brain tissue of model mice(P<0.01,compared with the model).2 the effect of compound 9714 on learning and memory ability of YD model rats induced by permanent bilateral carotid artery occlusion.The ability of learning and memory was significantly decreased after permanent bilateral carotid artery occlusion. Compared with the vehicle group, compound 9714 can improve the ability of learning and memory in model rats. It showed that in the Morris water maze test, compound 9714(10mg/kg, 20mg/kg, 40mg/kg) can shorten the escape latency time of the second and third days in place navigation test((P<0.05,P<0.01,compared with the model) and increase the percent of primary flat floor quadrant swimming distance to the overall swimming distance in spatial probe test in model rats(P<0.05, compared with the model).3 the effect of compound 9714 on blood brain barrier (BBB) permeability of communal carotid artery occlusion (CCAO) model rats.We examined Evans Blue's dye content in brain tissue in CCAO model rats, and found that after CCAO, the content of Evans Blue in brain tissue increased. Compound 9714(10mg/kgN 20mg/kg% 40mg/kg) can lessen the content of Evans Blue(P<0.05,P<0.01,compared with the model) in brain tissue in model rats by reducing permeability of BBB.
