Object: To study effect of recombinant human growth hormone(r-hGH) on cell cycle of a human breast cancer cell line MCF-7 , and study the effect of r-hGH plus 5-Fu on the effect of chemotherapy in MCF-7. Method: MCF-7 cells were cultured for 48h in RPMI-1640 with r-hGH for different concentrations and with 5-Fu and r-hGH for different concentrations ,then cells were gathered and evaluated for cell cycle by flow cytometry(FCM). Results: ①Comparing with the control group , r-hGH had no significant effect on the percentage of G0-G1, S and G2 phase of MCF-7 (P>0.05).②Addition of 5-Fu to the culture medium raised the percentage of G0-G1 phase significantly(P<0.05)and reduced S phase significantly (P<0.01) , amd did not change the percentage of G2~ M phase significantly(P>0.05).③Comparing with the control group , 5-Fu plus r-hGH raised of the percentage of G0-G1 phase significantly(P<0.01)and reduced S , G2~ M phase significantly (P<0.05). Conclusion: The r-hGH might not have effect on the cell cycle of MCF-7 cells. In vitro r-hGH might promote the sensitivity of chemotherapy with 5-Fu in MCF-7 cells...
