Objective: To probe the alterations of serum hyaluronic acid,hepatocellular apoptosis and apoptosis-regulating gene expression,regular liver function in rats with carbon tetrachloride-induced liver cirrhosis,the relationship between these indexes and liver reserve function after partial hepatectomy are also studied.Methods: 90 male Wistar rats randomly divided into 3 groups,9W,12W,15W were experimental groups;30 male Wistar rats randomly divided into 3 groups, were corresponding control groups,developing models of rat liver cirrhosis with complex factors (CCL4,alcohol),and the control group with saline.The blood and the liver tissue were taken for measures of the Group 9W,12W,15W after 9,12 ,15 weeks ,and those of the corresponding control group were killed at the same time.15W group was the partial hepatectomy group, The blood and the liver tissue were taken inpreoperative and postoperative for measures. The blood was taken for the determination of serum hyaluronic acid and regular hepatic function.The liver tissue were taken for pathologic examination,in situ TUNEL and Bcl-2,P53 by streptavidin peroxidase method.Results: Serum HA levels of the liver cirrhosis experimental groups were significantly higher than control group,9w(150.28±76.24ng/ml),12w(425.66±54.94ng/ml),15w(704.85±158.58ng/ml),serum HA levels of the partial hepatectomy were significantly higher than preoperative group,15%(937.81±375.2ng/ml),30%(1133.9±393.9ng/ml),45%(1329.0±148.1ng/ml)(P<0.01).Hepatocellular apoptosis in experimental groups was significantly higher than control group,9w(7.38±1.71),12w(14.81±2.99),15w(26.55±7.94).Hepatocellular apoptosis of the partial hepatectomy was also higher than preoperative group(26.55±7.94),15%(34.17±11.8),30%(44.57±15.49),45%(49.71±19.98)(P<0.01 or P<0.05). The indexes of hepatocellular apoptosis and serum HA level were positively correlated ,The indexes of hepatocellular apoptosis and serum HA level were also positively correlated with liver cirrhosis and partial hepatectomy .The indexes ofhepatocellular apoptosis and Bcl-2 were negatively correlated , The rates of hepatocellular apoptosis and p53 were positively correlated .Conclusion: The serum HA levels became higher after more aggravate of liver cirrhosis, The indexes of hepatocellular apoptosis and serum HA levels were positively correlated ,The levels of serum HA may be more accurate than regular hepatic function in predicting liver reserve function...
