Liver fibrosis is the pathological base of liver cirrhosis, which is one of the common diseases threatening human being's health. Liver fibrosis is a step in the pathological process of liver cirrhosis, developed from chronicle liver diseases caused by liver-damaging factors (such as hepatitis viruses). Slowing down or preventing liver fibrosis of chronicle hepatitis from developing will cure thousands of patients who are suffering from the diseases. Unfortunately, no dug has been found to date to cure liver fibrosis with highefficacy and low toxicity.For the last decade, significant progress has been made in the mechanismstudy of liver fibrosis. Liver fibrosis is a key step in chronicle liver diseases. In the initiating stage, factors such as viruses, liver damage and inflammation activate HSC, causing liver fibrosis to develop, leading to liver cirrhosis eventually. Therefore, treating liver fibrosis to cure chronicle hepatitis should target on the causes, protect liver cells and enhance patient's immunologicalabilities. These would prevent liver fibrosis and liver cirrhosis from forming.Chinese traditional medicine provides a great opportunity for the study of curing liver fibrosis caused by chronicle hepatitis. It has become one of the focal points in modern Chinese medicine pharmacology and liver disease research to search for effective drugs to cure liver fibrosis caused by chronicle hepatitis. Chinese medicine " SHUANGJIAWULING decoction / capsule " is formulated based on " WULINGWAN" modified with components for invigorate the circulation of blood make hardly become the softness and the knot stanza the dissipation according to Chinese medical principles. This study establishes cell and animal models and then compares current drugs for treating liver fibrosis with house-made "SHUANGJIAWULING decoction / capsule " . Furthermore, the cellular and molecular mechanism of treating liver fibrosis with multiplex Chinese traditional medicine preparation is confirmed and experimental data of reverting liver fibrosis are collected. The study provides powerful evidences for the future clinical applications of Chinese medicine.To study the mechanism of SHUANGJIAWULING decoctionin/ capsule curing liver fibrosis, the following study has been accomplished:1. A reliable and scientific animal model of liver fibrosis has been established. We have made our best effort to make the model close to the real human liver fibrosis in terms of pathological change and progression.2. Based on this animal model, the molecular mechanism of the degradation of liver fibrosis has been studied systematically at different angles and levels such as cell, protein, and gene expression. This isto understand the acting stages of the Chinese medicine and to collect data of treating liver fibrosis with Chinese medicine.3. Based on the theories of Chinese medicine/pharmacology and modern medicine about liver fibrosis, a brand new multiplex Chinese traditional medicine preparation has been formulated in the forms of decoction and capsule. The clinical and experimental studies have been performed using data from liver function, liver fibrosis, pathology and electronic microscope as benchmark with proper clinical and experimental controls.4. Based on the multi-clinical studies, an effective method for treating liver fibrosis has been established.Results and discussion1.Animal study has confirmed that SHUANGJIAWULING decoction/capsule hasthe ability to prevent and cure liver fibrosis. The possible mechanism may be:by inhibiting the activation of HSC and the inhibiting the formation of TIMP and promoting the degradation of ECM;by inhibiting the mRNA expression of TIMP-1, TIMP-2 and protein expression to reduce the inhibition to MMPs and to enhance the degradation of ECM;By accelerating the degradation of type-I and type-III collagens, and then preventing them from formation and precipitation.2.Clinical data have indicated that SHUANGJIAWULING decoction...
