The effect of apatite nanoparticles on cancer cells is a kind of biological effect of inorganic nanoparticles, and it had also been studied by the Biomedical Materials and Engineering Research Center of W.U.T since many years before.On the basis of the former research, the author selected'The effect of HAP nanoparticles on the hepatocellular carcinoma cell and its telomerase gene expression' as his main research content in the subject. By studying the telomerase activity, the purpose of this article is to investigate the anti-carcinoma effect and mechanism of apatite nanoparticles at the viewpoint of gene level ,and finally to offer some theory grounds for basic and clinical medicine. For the experiments, the Bel-7402 and L-02 were cultured in vitro.With the help of computer image analyse system, their characteristics and biological behaviors were observed. The Methyl thiazolyl tetrazolium (MTT) test usually used to evaluate the proliferation of cells which cultured in vitro because of its such advantages as high sensitivity, good repetition, fast and simple operation, and little influence by subjective factors. Extracorporeal cell culture and MTT colormetric assay were used to measurate the chemosenstivity of hepatoma cells to the HAP nanoparticles and five kinds of traditional antitumor drugs, such as cisplatin. doxorubicin, vincristine, mitomycin, cyclophosphamide. In order to determine the effect of HAP nanoparticles on the expression of telomerase gene,the telomerase in situ hybridization test kit was used and the result was contrast with that of a telomerase inhibitor(cisplatin).Then. conclusions are made: l.In the same operation times(72h), a majority of the traditional antitumor drugs with different concentration show goodresults of inhibiting the cancer cells in vitro. The inhibition rate of the HAP nanoparticles to the hepatoma cells is lower than that of the majority traditional antitumor drugs. However, it has little effect on the normal lever cells. It will have some advantages to the traditional antitumor drugs, if the HAP nanoparticles becomes a kind of antitumor drug some time in the future.2. The effect of HAP nanoparticles on the expression of telomerase gene:After treated ,the hepatocellular carcinoma Bel-7402 Cells with cisplatin and HAP nanoparticles at a high concentration for 4 hours respectively, the group treated with cisplatin is positive expression and the group treated with HAP nanoparticles is weak positive expression. Result of this preliminary experiment demonstrates that HAP nanoparticles has some potential influence on the expression of the hepatocellular carcinoma Bel-7402 cell telomerase gene .
