Objective To evaluate the effect of hTGF-β1 gene on the cell apoptosis and its control proteins Bcl-2, Bax in nucleus pulposus, so as to activate the cells and prevent the tissue from degeneration. Methods All of 15 adult rabbits were used. The transforming growth factor-β1 (hT-GF-β1)as a exogenous gene was injected directly into L3-4 intervertebral space of 5 adult rabbits, and another 5 adult rabbits had injected PBS into the L4-5 intervertebral space as the controlling test group. The last 5 adult rabbits had been the normal group. The corresponding discs were removed 3 weeks later, and the apoptosis cells were examined and analyzed using TUNEL the proteins Bcl-2, Bax were analyzed by immunohistochemistry stain and Vidas analysis system. Results The morbidity of apoptosis cells in the injecting hTGF-β1 gene group is less than that in the another two groups . Also, the proteins Bcl-2 expressed more morbidity, the proteins Bax expressed lesser morbidity. There is significant different of OPTDM after compared by the one way analysis of variance. Conclusion Transforming the hTGF-β1 gene could lessen the cells with occurrence of apoptosis significantly by enhancing the expression of the protein Bcl-2 and depressing the expression of protein Bax.
