Objective: To evaluate the effect of inositol hexaphosphate (IP6 or InspG) on 1, 2-dimethylhydrazine (DMH) induced colon carcinogenesis in Wistar rats and discuss the mechanism underlying the anti-neoplastic potential of IP6. Methods: 30 four-week-old male Wistar rats were divided randomly into 2 groups of 14 rats each and fed with diets based on AIN-93G, the treatment group were given 2% Na-InsP6 supplemented water, the control group received tap water. Four weeks following the beginning of their special diets, the rats were given a weekly injection of DMH (20mg/kg) for 20 weeks. The rats were killed after 26 weeks. The number of total macroscopically visible neoplasms was counted and the volume of individual neoplasmas was calculated. Proliferation cell nuclear antigen (PCNA) and SOD, GSH-Px and MDA of colon were analyzed as well as Natural killer cell activity.Results: 1. The incidence of tumors in Wistar rats treated with DMH without InsP6 was 79%(ll/14) compared to 89%(8/9) in rats treated with DMH plus InsP6. But there was no significant difference between the two groups(P>0.05). However InsP6 significantly reduced the number of tumors per rats 73% (t=3. 056, P<0.01) , tumor volume by 83% (t=2. 291,P<0.05).2. Treatment of rats with Insp6 significantly raised SOD and GSH-Px activity of colon (SOD: t=9.014, P<0.01; GSH-Px: t=13.264, P< 0.01) and reduced MDA content (t=18.608, P<0.01).3. The expression of proliferation marker PCNA was significantly down-regulated (t=2.680, P<0.01) by InsP6.4. Treatment of rats with InsP6 significantly enhanced NK activity (t=16. 032, P<0.05).Conclusion: A treatment regimen of 2% Na-InsP6 in drinking water was effective in significantly reducing the risk of large intestinal cancer in Wistar rats. We postulate that InsP6 may exert its antineoplastic effect by way of its antioxidant function and lowering cellular proliferation as well as enhancing the NK cell activity.Zhang Haiping(Nutrition And Food Hygiene) Directed by Song Yang...
