| Objective:The main objectives of the present study are:1To research the function of prevention and cure and the possible mechanism that Liansu antiemetic capsule(LAC) influence delayed emesis due to chemotherapy.2 To research some relations of emesis due to chemotherapy between the "huoreneiyu" morbid mechanism of Traditional Chinese Medicine(TCM) and the mechanism of modern medicine.Methods:Experiment 1:The improving of pigeon's vomiting model by the chemical medicine cisplatinThe first step: 50 health pigeons weighted 450~520g were randomly divided into 5 groups that given different dose of Cisplatin(20mg·kg-1,16mg·kg-1,8mg·kg-1,4mg·kg-1,3mg·kg-1). We injected the chemical medicine Cisplatin in pigeon's abdomen every 24 hours, then,let these pigeons freely drink dextrose water of 10% in order to maintaining their nourishment.Then,we observed the pigeons' vomitting condition in 8 hours.The second step: Selecting 10 health pigeons,we changed the injecting dose of Cisplatin according as the first step'sresult that we observed,that's to say, injecting three days with the dose of 4-4-3mg·kg-1(the first day use 4 mg·kg-1, the second day use 4 mg·kg-1, the third day use 3 mg·kg-1, the same next).then we continued the experiment. The third step: We continued improving the injecting dose of Cisplatin according as the second step's result that we observed,that's to say, injecting three days with the dose of 4-3-3mg·kg-1.then we continued the experiment.Experiment 2:Liansu antiemetic capsule's influence on the pigeons' vomiting times,gastrointestinal hormones and duodenal mucosa after chemotherapy with CisplatinFeeding the pigeons a week normally, then we divided them into 5 groups randomly,those were blank group(BG),model group(MG), ondansetron group(OG), the low dose group of Liansu antiemetic capsule (LLAC,360 mg raw drug /kg), the high dose group of Liansu antiemetic capsule (HLAC, 720mg raw drug /kg).Each group had 8 pigeons.We began giving drug to BG 3 days before making model and to OG 1 day before making model, according as the characteristic of western medicine and Chinese traditional medicine while being used in anti-vomiting due to chemotherapy in clinic.In order to having the same stimulus,the pigeons were filled the same proportion saline when have no drug to fill,all with the capacity of 4ml/kg/ d and once a day.The fourth day,we began making model conculting literature.Every group were injected in abdomen with the capacity of 2ml/kg/ d,once a day,total 3 days. The BG were injected saline and the MG,OG, LLAC, HLAC were all injected Cisplatin.In the three days,the dose was 4mg/kg/d,3mg/kg/d,3mg/kg/d in turn.Every group continued filling medicine or saline in stomach in the three days when we made model. Then we observed and noted the pigeons' vomiting latency (the time between injecting Cisplatin in abdomen and occurring the first emesis) of the first day,the vomiting times in 8 hours every day after injecting Cisplatin.Every group were fasted 3 hours when we made model 3 day later,then decollated the pigeons' head in order to gaining blood and collected duodenal specimen of light lens.Each pigeon was gathered 4 ml and 2 ml blood.Isolated serum from the 4 ml blood(4℃ isolated 15min,3500rpm). The serums were saved not higher than -20℃ in order to determining Epidermal growth factor(EGF) and Gastrins(Gas).Each 2 ml blood was put in a test tube that was chilled by ice and had been put in 10%EDTA.Na2 30ul and Aprotinin 1000KIU,then isolated plasma in low temperature centrifuges for 15min, 1500rpm. All the plasma were saved in -20℃ in order to determining Vasoactive intestinal peptide(VIP).Each pigeon was dissected abdomen.We selected a piece of duodenum,2cm long,and washed it with saline,then we fixed the specimen in 10%formaldehyde over 24 hours.Experiment 3:The influence on rabbits' gastric motility of Liansu antiemetic capsule 40 healthy rabbits,including male and female,weighted 1.8~2.2kg were randomly divided... |
