| ObjectiveTo investigate the effect of borneol on neurotransmitter and vasoactive substance in rats plasma and hypothalamus, we study the relations of dosage-effect and time-effect, for probing into the possible functionary mechanisms and target of borneol .Methods168 wistar rats were randomly divided into 5 groups, given 0.9%NS, 1%CMC-Na, and three different dosages of nature borneol two times every day. At 0min, 45min, 24hr, 72hr, 120hr, monoamine neurotransmitter levels in rats plasma and hypothalamus were measured by HPLC combined with electrochemical detector method, and monoamine oxidase, ACh in brain tissue were determined too. The plasma contents of ET,ANP, CGRP, Angâ…¡ were determined by radio immunoassay and NO by method of nitrate reductase. Results1. In plasma, after given borneol 45 minutes, the levels of NE were decreased. The differences of middle-dosage and low-dosage groups are significant compared with 1%CMC-Na group (P<0.05). After continuously given 24 hours, borneol can reduce the contents of NE in plasma too. The differences of middle-dosage and high-dosage groups are significant compared with 1%CMC-Na group (P<0.05).The low dosage borneol also showed slight decrease, but the high dosage is the best. At meantime the borneol can reduce the E levels in plasma after given 45 minutes, but the difference is not significant (P>0.05). Continuously given 24hours, borneol can reduce E contents in plasma. The contents of E in middle-dosage group decreased more significantly than that in the 1%CMC-Na group (P<0.05). The contents of ET in middle and high group decreased more significantly than that in the 1%CMC-Na group (P<0.05).It did not have a significant effect on NO and CGRP contents in plasma, but the contents of NOS were increased. The experiment on effect of borneol on renin-angiotensin system shows:It did not have a significant effect on ANP, but it has a slight decrease on Angâ…¡.2. In hypothalamus, comparing with 1%CMC-Na group we find that the middle dosage and high dosage borneol can decrease markedly the NE and E levels in hypothalamus after given borneol 45 minutes (P<0.05), and after continuously given borneol, the E and NE contents in hypothalamus are reduced too. After given borneol, the effect on 5-HT contents in hypothalamus has not been found, compared with 1%CMC-Na group. after given borneol five days, the middle and high dosage can markedly decrease the DA levels in hypothalamus. On ACh contents of brain tissue, the difference is not significant compared with borneol groups and 1%CMC-Na group,while monoamine oxidase is increased by borneol.3. The changes of monoamine neurotransmitter in rats plasma and hypothalamus are similar.ConclusionThe above results suggest that borneol can decrease the contents of catecholamines and resist excitement of sympatho-adrenergic system. It also can reduce the ET levels obviously and decrease Angâ…¡slightly in plasma. In a word, borneol can regulate the function of physiology and pathology by effecting neuroendocrine,and this may be one of mechanisms on prevention and treatment of cordial diseases. |
