Purpose:This research is to investigate the effect of Chinese compound medicine Kang Xian Zeng Zhi (KXZZ) Pellet on the activity of NMDA receptor in brain of kinding model rats and their studying memory in water maze induced by pentylenetetrazole(PTZ).Methods:The rats were randomized into 4 groups: model group, western medicine group, KXZZ high dosage group and KXZZ low dosage group, 8 normal rats were selected as the control. Acute seizure of the kinding model was induced by PTZ with the dosage of 32mg/kg, and the rats at V,VI degree were selected as the objective of study. Observation was as follows: Firstly, the objective of study of behavior was performed. Secondly, the studying memory ability was tested. Finally, the activity of NMDA receptor of cortex and hippocampus was measured.Results:1. The behavioral study showed that KXZZ pellet could effectively inhibit the seizures induced by PTZ. It had significant difference compared with the model group in the numbers of rats with seizures, degree of seizures and latent period (P<0.05), and it's function was related with the dosage. There was no significant difference among the three treatment groups.2. The test of the activity of NMDA receptor displayed that: There was notable difference between the normal group and the model group (P<0.05), the activity of NMDA receptor of the model group in cortex and hippocampus was remarkably increased. The three treatment groups had no significant difference in hippocampus compared with the model group, although their activity of NMDA receptor was decreased. There are significant difference in cortex between KXZZ high dosage group. Western medicinegroup and the model group. The activity of NMDA receptor was decreased notably (P<0.05). The low dosage KXZZ group had no notable difference in cortex compared with the model group although could decrease the activity of NMDA receptor.3.The test of water maze showed that: There was notable difference between the normal group and the model group (P<0.05), the time of the model group rats seeking the platform was extended. Compared with the model group, the time of KXZZ high dosage group rats seeking the platform was shortened remarkably(P<0.05), but there was no significant difference between the low dosage KXZZ group, western medicine group and the model group, although their time was shortened too.Conclusion:Chinese medicine of KXZZ pellet could effedctively inhibt the seizures induced by PTZ, reduce the degree of seizures and prolong the latent period. It also can improve the ability of studying memory of the kinding model rats. It's function may be related with regulating the activity of NMDA receptor.
