[Objective]:To investigate the variety of hTERT ( human telomerase reverse transcripase) in the course of glioma'(astrocytoma, medulloblastoma, oligodendroglioma,ependymoma) development ,the possibility of using as it an valuable tool for easily diagnosis or precise prognosis of gliomas.[Methods] : Human brain specimens from 45 gliomas with differential subtypes and grades and 8 normal brain tissues,were examined for hTERTmRNA by method of ISH(in situ hybridization).While PCNA protein were detected by immunohistochemistry method (S-P) in above spicemens.[Results] : Human brain specimens from 45 gliomas with differential subtypes and grades were devided into two groupsilow malignant group(LMG)which concludes 16 cases of 2 astrocytomas, 9 grade II astrocytomas, 2 oligodendrogliomas,4 ependymomas; high malignant group(HMG) which concludes 13 cases of9 anaplastic astrocytomas, 4 anaplastic ependymomas; 16 cases of 6 medulloblastomas, 10 glioblastoma multiformes; The posstivity of hTERT was 60.0%,31.25%,75.90% in total group, LMG and HMG retrospectively.No hTERT was detected in normal brain tissues.Through stastics test,there exists remarkable difference between these groups (P<0.05) . The posstivity of PCNA in LMG, and HMG are 8.50±1.75%,32.00±6.35% retrospectively. The correlation index between the expression of hTERT and the posstivity of PCNAL1 is r=0.727 (P<0.05) .[Conclusion] :Through hTERT's expression was not detected in normal brain tissues,it could be tested highly in gliomas with an increasing trend.It shows that hTERT may occur in the progress of gliomacarcinogenesis and play an important role.hTERT may be a useful new marker in diognosis and prognosis of glioma.
