Objective To observe the effect of different concentration of astragalus membranaceus on nitric oxide (NO) level of human vascular endothelial cells cultured with high glucose and free fatty acids (FFA) for 24 hours. Methods Cultured human vascular endothelial cells were used for experiment. NO was measured by means of nitrate reductase.Results The levels of NO in the high glucose concentration (Glu30mmol/L) group and Glu30mmol/L + palmitate (PA) 0.5 mmol/L group were significantly higher than that of control group (P<0.01). After treating with astragalus membranaceus the levels of NO were decreased. In contrast, the level of NO in the supernatant of oleate (OA) 0.5mmol/l group, PA 0.5 mmol/L+OA 0.5 mmol/L and Glu 30mmoI/L +OA 0.5 mmol/L +PA 0.5 mmol/L group were significantly lower than that of control group (PO.01). After treating with astragalus membranaceus the levels of NO were increased.Conclusion This study indicated that high concentration of glucose increased NO production, whereas high concentration FFA inhibited NO production in endothelial cells 24 hours, astragalus membranaceus could improve the abnormal levels of NO. It might be one of the essential mechanisms of astragalus membranaceus in treating diabetic nephropathy...
