AIM: Investigation the relationship between differentiation of osteoblasts and bFGF (basic fibroblast growth factor) regulation of osteoblastic activity and apoptosis. Materils and method: Obtaining experimental cellular model by isolating and culturing in vitro of osteoblasts from SD rats' fetal calvaria . Differentiation stages were divided . Three days after bFGF (10μg/L) had been put in medium culturing different stages of differentiation , osteoblastic activity in different stages of differentiation and effects of bFGF on osteoblastic activity were determineed by means of biochemistry and radio-immunity techology . The contents of DNA and expressions of Bcl-2 and Fas were tested by FCM (flow cytometer ) . The effects on osteoblastic apoptosis in different stages of differentiation and related gene Bcl-2 and Fas by bFGF were analyzed . The group without bFGF were used as control group. Result : The experimental cells obtained through isolation and culture from SD rats' fetal calvaria have been proved to be highly purified osteoblast by morphology and determine of ALP (alkaline phosphatas),OC(osteocalcin). These experimental cells are charaterized by differentiation stages . The ALP content in bFGF groupwas lower than the ALP content in control group . The OC content in bFGF group had on increasing tendency compared with OC content in control group . According to the testing of FCM, the rates of osteoblastic apoptosis during different differentiation stages in bFGF group were lower than the rates in control group. The expressions of Bcl-2 in bFGF group were higher than the expressions of Bcl-2 in the control group. The expressions of Fas were signficantial lower than expressions of Fas in control group . Conclusion: bFGF inhibits differentiating osteoblastil ALP activity, but it is not significan that the effects on the increasing of OC by bFGF. bFGF inhibited the osteoblastic apoptosis in differentiating and the inhibition mechanism was associated with the regulation of expressions of Bcl-2 and Fas .
