Studies on antineoplastic active components in Ardisia crenata SimsAt present, malignant tumor is one of the most serious diseases that threaten human beings. The characteristics of malignant tumor is abnormal cells lose growth control, and they diffuse from original location to other locations. If the cell proliferation beyond control, the human main organs will be injured, eventually death will be the result. Currently, world-wide activities in cancer research, prevention and cure are increasing.It is greatly progressing in cancer prevention and cure by Chinese medicine. It is demonstrated not only by the clinic curative effects, diverse ways of diagnosis and treatment, but also by progressing in the academic research. Chinese doctors are exploring the traditional Chinese medicine with up-to-date science knowledge, and have elucidated some of the treatment mechanisms by modern theories, and have developed the cancer treatment by Chinese medicine to a new stage.At present, the hot spot is the research of antineoplastic effects by extracting effective components from Chinese herbs. It is improved that Chinese herbs effective components have many characteristics including high effect, low toxicity, diverse approach, and diversity. It is an urgent task to develop effective components from Chinese herbs to treat cancer.Ardisia crenata Sims is one of Chinese herbs in Zhejiang, Jiangxi, Guizhou in China. We extract active components from Ardisia crenata Sims with clear antineoplastic activity through many years of efforts.This paper is a part of antineoplastic activity research of Ardisia crenata Sims. In the past years, we isolated many components from Ardisia crenata Sims, including saponins, coumarin, bergenin, nor-bergenin, 11-o-syringicacaoly bergenin, rapanone, β-sitosterol, daucosterol, etc. Pharmacological studies showed that ardisiacrispin A, ardisiacrispin B, ardiscrenoside A and ardiscrennoside B in Ardisia crenata Sims were the components with antineoplastic effects. In this paper, antineoplastic action of saponins of Ardisia crenata Sims (ZSG-80) is investigated, and the treating mechanism is explored. The paper includes two experiments: antineoplastic experiment and analysis of its mechanism. Antineoplastic experiments are mainly researches of ZSG-80 effect on transplantation lung tumors and colon cancer in the nude mice, and inhibition action to in vitro cell growing, such as PG, Bel-7402, KB, HCT, HELA, HL-60 etc. analysis of mechanism include two aspects: the cytology research on proliferation of human carcinomata of PG, Bel-7402, HL-60 in vitro; and the influence on mice cell immunity. 1. Effect of ZSG-80 on the model of transplantation tumors in nude miceUsing the model of transplantation lung and colon tumors in nude mice, we observed 40mg/kg ZSG-80 have significant antineoplastic effects and innocuity。2,Effect of ZSG-80 on the tumors cell proliferationThe effect of ZSG-80 on the tumors cell proliferation was observed. In the in vitro experiments of cell culture, ZSG-80 showed significant inhibition effects on proliferation of human carcinomata of PG, Bel-7402, KB, HCT, HELA and HL-60.3. Analysis of ZSG-80 antineoplastic mechanism experimentProliferation of human carcinomata of PG, Bel-7402, and HL-60 in vitro was used. The mechanism of action was preliminarily elucidated, including inhibition on proliferation, down-regulation of clone ability, inhibition on activities of neoplastic cell, direct attack on the cell, disturbances in the cell cycle and induction for cell apoptosis. 4. Effect of ZSG-80 on the mice cell immunityUsing MTT method, we showed ZSG-80 have no significant effect on the level of mice cell immunity.Based on all the above researches, the author's conclusions are as follows: ZSG-80 has significant antineoplastic effects to mouse transplantation tumors, such as Lewis, H22, U14 and S180.
