Loss of tissue or organ is one of the most important and extensive problems of human ealth.Currently,the solutions for this problem are transplantation,reconstructive surgery,or the use of mechanical devices.Reconstruction may lead to long-term problems.Transplantation depends on the doners whose number is quite restricted. To solve these problems,a new field called tissue engineering has emerged. Tissue engineering is an interdisciplinary field that applies the prinples of engineering and the life maintain or improve tissue function. After that,as a series of techniques are being developed for culturing and reconstructing a variety of tissues both in vitro and vivol,it is possible to fabricate dile duct model in vitro whose struture and function are similar to autogenous duct. In my experiment, VSMCs obtained from the media of human embry arterial were cultured successfully ,and the Cells were attach to PLGA and grew favorably on the surface of it.we found that PLGA was a suitable cells carrier,cells and PLGA complex could be a suitable of media of dile duct. Part IObjective:To detect the approaches to culture the human embryo vascular smooth muscle cells(VSMCs) in vitro andcryopreservation and resuscitation. Methods:VSMCs obtained from the media of human embryo arterial were cultured in vitro and passed Stimes, then identified and cryopreserved. The frozen cells were resuscitated after two weeks, 1 months, 2 months and 3 months. Results:Cultured VSMCs were obtained .The frozen cells were resuscitated successfully.The resuscitated rates were more than 80%. Conclusions:The establishment of VSMCs culture in vitro and its cryopreservation , resuscitation, made the VSMCs culture system more perfect, which would be helpful to the studies of drugs for the cardiovaslar dieases and the research on the tissue engineering dile duct...
