Objective : To investigate the expression and significance of matrix metalloproteinase-2, 9 and transforming growth factor- β1 in the vocal polyps, and to further study their role in the pathogenesis of the vocal polyps. Methods: 1 Hematoxylin and Eosin(H&E) staining and Grocott-zones PISM staining was used to study the change of Basement Membrane Zone(BMZ) in the vocal polyps compared with the normal human vocal folds and nodules . 2 The expression and distribution of TGF- β1, MMP-2 and MMP-9 was detected in the vocal polyps nodules and the normal human vocal folds by imrnunohistochemical method Results 1 Compared with the normal human vocal folds ,the vocal polyps commonly exhibit an unaltered BMZ width of surface epithelium , but the BMZ of the neovasculature of the vocal polyps stained significantly thiner or weakerthan those of the normal human vocal folds(P<0.05) ,associated with edema degeneration exangia thrombosis of interstitial substance of Lamina Propria . The Vocal nodules commonly have a significantly thickened BMZ of surface epithelium compared with the vocal polyps and the normal human vocal folds(P<0.05) , but the BMZ of the neovasculature of the vocal nodules was as same as those of . the normal human vocal folds . 2 MMP-2 MMP-9 and TGF-β1 was significantly over expressed in the vocal polyps, Compared to the vocal nodules (p<0.05) and the normal human vocal folds (p<0.05) 3 The expression of MMP-2 MMP-9 and TGF-β1 in the vocal nodules was unsignificantly lower than the normal human vocal folds (p>0.05) 4 There was correlation between the expression of MMP-2 MMP-9 and TGF-β1 in the vocal polyps (P<0.01)Conclusion 1 Thinness or weakening in the vocal polyps may be critical factor of the vocal polyp formation 2 MMP-2 and MMP-9 were strongly conflation with Thinness or weakening of the BMZ , they seem to be involve in the degradation BMZ and cause pathological changes in the vocal polyps 3 TGF- β1 may play inhibitory action in vocal polyps formation.
